No growth on agar? Day 7

This is my first run using agar. I ordered some PDA online, mix it according to the directions. I used a syringe of B+ that I've been sitting around a while, so there's already some mycellium in the syringe. I place 1 2 3 drops on several glad mini rounds. There seems to be condensation on the walls and a tiny pool of water in my dishes. I opened one up just to be sure and so nothing, not even to contaminate. Any suggestions?

Well I guess the silver lining would be they haven't turned green.
As for advice I dunno really I poured agar for first time yesterday. Hope they dont take 7 days tho :p. Oh you did put them in a warm place right?

how do u know there is mycelium in the spore syringe?
after u flame the needle on ur syringe, lift the agar lid, and cool the needle on the surface of the agar by digging into it and scratching it. in those divots and scratches u made, that is where u should drop ur spores if possible.

Be patient. 7 days is nothing. If you don't see growth by day 14 then I would suggest starting over. If you do decide to start over, use less agar so that it makes a softer surface for the spores to germinate on.

@mike - yea I furked up there too.. saw this tek that said to use 11.75g per 250ml. Then I read ppl use as little as 7g or less

I would still be patient because I have had no problem germinating spores on stiff agar. It usually just takes a little longer.

i use stiff agar (preference, but it makes transfers easier IMO), so i agree with mike, it will just take a little longer vs. softer agar.
but if u dig some grooves into the plate with ur needle after u flame it red hot, and shoot the spores into those grooves, it will help germination by keeping those spores moist and surrounded.

I used 10g/100ml, so 7/100 would work as well?

I just did my first agar dishes 2 days ago and nothing so far so yea i think it takes a bit longer. I have a small amount of water in one dish so i tilted it to get it to one side , is there any hope for this dishes like this?

Yeah there is hope. The water can make bacteria spread like a mother fucker if it presents, but water on an agar surface is far from a deal breaker.
