Flow hood alternatives?
    #22337598 - 10/05/15 04:39 PM (8 years, 3 months ago)

Just getting back into mycology work and with this I jumped into agar/spawn bags. As I'm reading more I'm hearing the stress doing agar work w/o a flowhood causes. I would like to make something basic for the time being untill I have the time and funds to invest in a flowhood. My question is: what can I make that has a better sucsess rate than a glovebox without having to build a FH.
I was thinking something along g the li E of a hepa pushing into a tote.

a still air box. I think there's a link for how to build one in my sig.

technically they are better than a glove box. and you can't get much cheaper than a SAB (still air box)

Yup you can achieve a pretty decent success rate with a SAB, mine was a bit restricting in movement and got on my nerves a bit. I much prefer working with a flowhood. They key to success is also probably more so with sterile technique than with whatever you are working in.

^ This. Totally.

I just did what scientists do at the bench. If you take a Bunsen burner and work in back of the flame (without burning yourself, lol) the heat will draw in all the bad air. If you are working with agar you have to get used to opening the plates in the front and not the back (the side closest to the flame; open it as if it were Pacman).

Easier than a SAB in terms of construction but harder to get used to. If you can, you'll probably have more luck will a properly cleaned still air box.

Edit: Found this link, may be of some assistance:

Micro: I was under the impression that useing a flame like that to keep an area clean doesn't work. Any recent discussions you might be able to point me towards?

That SAB is badass and so easy...  I will be doing that!  I was getting a little irritated at some of the flow hood designs because they are expensive bulky and not very stealthy but a SAB is very stealthy,  cheap,  simple,  and the idea seems very effective.  A flame inside a SAB may not be the best idea right lol =p Thanks for sharing the design. -J*

instead of a flame try isopropyl alcohol and/or oust.

iso alcohol is a poor substitute for flame sterilization. most people on here will tell you that.

I just keep a propane torch lit on a table near my SAB. whenever I need to flame I simply pull said item out of the SAB and apply the flame.

Just gotta keep safety in mind.

Still air box works, I just have an air purifying tower next to my work area. I keep my utensils in a uv-sterilizer cabinet when I need to set them down. I've been box-free for a while now and I haven't had any troubles.

Micro, that's a pretty interesting link you posted. It never occurred to me that the air circulation from the flame could create a sterile radius.
