Hello! This is my first time growing. A month ago I had one last dried shroom that I had and wanted to find a way to clone it because I live in CA and cannot order prints. I bought agar plates from Amazon and cut out pieces of dry tissue and used gills to use on the agar. I did this all in a SAB about a month ago I have an old post when I first put them on agar. I decided to transfer some small samples from the growth I had to new one and also decided just to try to put them already on some BRF jars I had ready. I just cut out wedges and place the onto the brf jar. I used about 4 wedges for each 4 jars I had to experiment. I used no dry vermiculite layer and only have to holes on each jar with foil over them. This is my results. The dry shroom is the one in my avatar. Pics starting from agar to jars. With only one transfer each. First transfer got moist and spread the spores everywhere but still good.

Interesting.
I haven't actually seen an Agar to BRF attempt.

Hopefully it doesn't stall before getting to the bottom.
You could also try to get an invitro fruit and spore print that to make a syringe for a standard BRF tek.

What do you mean stall? Does this happen commonly and will it stop growing. I was hoping on getting at least one mushroom off this attempt I didn't think I would have gotten this far as many people said I wouldn't even get any growth other then contams when I first did the agar attempt. Before I place the wedges in the jars I cut out small samples for new agar plates that look way better then the first time. Am I luck or did I get blessed with the shrooms?

Looks pretty good to me.
It could contam at the bottom, it depends how fast it can colonize.

But if you have it colonize more than half of that, you can just scrape away the uncolonized stuff with a spoon, then fruit the colonized sub you have.

When you inject spores you do it to 4 sides, so it can colonize quickly, but using agar skips your germination period so I'd say you have a decent chance at growing a few shrooms.

Then you'll have spores, and you can continue.

Dude! An ambitious undertaking for a first grow man!! Nice work!!!

Jars look clean and healthy so far.

BRF is a little bit forgiving as far as contams go so I think you will be fine. Ive only put agar to brf once and they took a little while to colonize but once it gets going it goes pretty quick

Nice work!

Thank you guys! Yes this is my first time doing any of this. A lot of people said I wouldn't even get growth from a dried shroom.  Yes i am very lucky with all of this good process I was desperatly trying to grow these so I did a lot of research to get these going. I am very lucky I diyeven get any contams when I grew these in agar and that I don't even have a dry vermiculite layer to block or contams

Have you more photos of the jars, are you making PF cakes? Have you open holes in the lid? or a filter?

I see tinfoil on your lids. Usually if you make a PF cake it has 4 holes in the lid, and a dry layer of vermiculite at the top, which acts as the filter. The tinfoil is usually removed as the verm is the filter and tight foil will prevent air exchange.

However if you put agar in at the top this layer is comprimised, so I would not take the foil off, if you do have 4 open holes it could be carefully loosened but not removed. So some air exchange could take place.

I have made BRF jars before with no top layer, they were only 1/4 filled and I was able to shake them to spread growth around, not as easily as grains but it mixed a bit. I thought of adding a large ball bearing to more easily shake/break it up.

I'll take more photos of the 4 jars tomorrow. They have two holes each with no dry vermiculite layers. No filters just foil I have not removed after inoculation in a SAB. These were just experiments I have done and wanted to see if people thought they looked alright. One of the jars looks a little pink on one side it has no mycelium growing on it yet, is this normal for vermiculite/ brown rice cakes to turn if they were over cooked or is this pink mold? I'll post a shot tomorrow.

Oh yea man if you are using agar to brf cake you need to skip the dry verm layer, but add a GE filter such as a piece of tyvek or a synthetic filter disc between the lid with holes and the metal band. Otherwise you are asking for contams

Here's a pic from agar to PF jars I did



I just have the foil has a filter. I know I'm asking for trouble but I heard bacteria can't grow up into the lid in the holes. I know I have to still worry about air Bourne contaminations. What happens to the old agar after its fully colonized? And when the top get covered in mycelium does that block out the co tams? Here are two pics each of the 4 growing update. The last two are the ones that has a pinkish spot on half the jar.

These are the two pics of the one jar that I have that has a pinkish discoloration on half of the jar. I have no seen it getting any brighter or any change that I can see from a week ago but looks to have spread. Is this just from not mixing the brown rice flour right?

I have seen a few threads that address similar coloration issues. Each turn purple after a few days which is a type of mold. Since I have not done cakes before, I can't be absolutely certain but it looks like mold.

You can take a marker and draw a boarder around the pink discoloration on the jar to see if it extends beyond the boarders.

Congratulations on getting such nice agar plates from that one dried shroom. Bravo!  As for the BRF--too bad you didn't do a transfer to rye berries. I have never done BRF, so can't advise you--but I think BRF was designed with spore solution inoculation in mind.

Looks like bacteria also.
If it colonized a good part of the BRF, you can scrape away contams under running water and use the rest.

Check my sig, I used some fucked up cakes and managed to get a decent monotub running with all of them.

Stop experimenting half way through your first grow. Grow it to maturity, watch it, learn from it. Do this a few times then experiment with different ideas and figure out what works best for you.

Thanks a lot! There is only one jar out of the 4 that looks bad. Should I just hang on to it and wait to see if mushrooms form? Would the mycelium cover the contam and fight it off or will it absorb it? The other jars are almost reaching the bottom I think in a week they will be full colonized. One other jar is taking a long time to cover but I see it still growing is that a problem?

Hell yeah dude, glad to see it's working out. Idk if you'd wana check this out it may help to get the cars colonized quicker than just dropping an agar wedge, http://www.shroomery.org/forums/showflat.php/Number/20429745 i cant speak from experience yet though, decided to hold off on trying to clone my old dry shrooms and order spores from a free spore company

I have never seen mushroom mycelium successfully take over a contam. By successful I mean cover it up and keep it from spreading.
I would let them all grow until fully colonized. There is a chance that you can separate the good from the bad on your questionable cake. But that is a desperate move.

You could attempt to make a liquid inoculate if you have a petri dish that is in good shape. I like using big syringes but a ten cc would do. Get a sterile syringe, sterile water, sterile needle 16-18g. Fill syringe with water. Crack open the petri. Squirt a few drops of water onto some good looking mycelium. Use the needle to cut up the mycelium, sort of stir it up with the water. Then suck it back into the syringe. You should see dozens of pieces of mycelium floating in the syringe. And there will be a lot more that you can't see.  Keep in mind that this whole process would be done in a glove box/sab/flow hood.

Really? It's kind of common for mycelium to do that in BRF. It can grow around and stop a contam, sometimes cover it up entirely.
If I ever get a contam that is small, I just scrape away the bad stuff with a spoon under running water.
Cheese grate and spawn to coir.
