Question about plates that get trich. Do you really toss the whole plate in the trash, or can i just drop the agar into the trash, wash the plate, wipe with bleach/iso, wash again. and reuse the plate?

Plastic petris are sterile from the manufacturer.
If you use it, you have to toss those after 'cause they'll no longer be sterile. It sounds like you need to tighten up your technique. Maybe when you're pouring them or storing them you're making a mistake or you're transferring sloppy. I only get trich on my last few plates after a long night of transferring and it's because I get sloppy.

I have pasty plates that have been run 100+ times. Many of them have had mode and bacteria in them at one point or another. I don't even wash them between uses. Just dump, wipe and they are ready to go.

I seriously love these things. The best part is being able to remove the aluminum foil easily and re-use it between PC cycles. If it rips it's whatever, I'll make a new one, but that extra dollar (Not to mention 20 minutes) saved from not cutting new foil every PC cycle.

Okay, seriously? How? My foil deteriorates and becomes shitty after like 2-3 runs. I've used thick foil, thin foil, doesn't seem to matter, it starts getting that shit all over the jars that makes them foggy, and the inside of the PC as well.

It's not really a calcium layer is it? Is isopropyl the best thing to use for it? I always thought you were supposed to use vinegar for some reason.
I dunno if it works with AAs being as they're made of aluminum but my old PC said to add a splash of white vinegar to the PC water to keep that film from sticking to the jars. That wouldn't fuck up an AA would it?
I'll spritz some iso on the jars and see if that works any better for now.

yup, vinegar (i use apple cider vinegar) in the water works and yes thats hard water calcium, I have it on everything.
it doesnt hurt anything beside visibility
AAs are tanks, I dont know how to hurt one
Ive used all 3 pcs, cheap china made prestos, AA pressure cookers and AA sterilizers/autoclaves.
they all work/maintenance the same imo.

Oh, that's a good tip, 13buds. Mine usually last about 10-15 cycles through the PC. Going to try your trick and see if I can get longer use out of them.

Ohhh that's right it's from the water, not the foil. Well, I'm betting some of it is from the foil, the foil DOES deteriorate and form weak spots and holes. But yeah that makes a lot of sense, I somehow forgot there was water being reduced in there. I never notice much of a water level change so it escaped me that that's happening every time.
Maybe it's just the shitty foil they sell around here.

So vinegar in the AA, no foul right?
not at all, it (vinegar smell) rinses out with hot water. plus it leaves it shiny clean.
I put all my store bought veggies and fruits in my pc and fill it up with water and Apple cider vinegar to clean off the chems that are on them. never had any issues afer several years of doing so.
evoo to lube the lid seal and vinegar on the inside if needed.

Why apple cider rather than white? How much are you using in a PC run? I only ever literally do a splash, doesn't always keep the shit off the jars.

Caustic soda/drain cleaner/sodium hydroxide should do the trick.
Most household bleach has sodium hydroxide added to it so I would be cautious about using it, this would be more dilute so might be OK.

They should do the same job. White is usually cheaper.

Boomertown said: I am about to give up on agar.....i don't understand how to get rid of said "mold" the fluffy white shit. I've done many transfers now and to no avail. Now I seem to be getting trich in half of the plates I do and the other half just suck. I use a SAB...I wipe down my plates with iso. I flame my scalpel after each transfer. I have my plates upside down. I always transfer from the best looking outside edge....I just don't get what I am doing wrong.
Have you ever tried to transfer that fluffy white stuff? Putting multispore to agar (especially high nutrient agar) results in fluffy myc. Usually my first plates are fluffy, then transfer to fresh plates that lack a little in nutrients. After 2 transfers and 2 recipe alterations I can usually come out with clean rhizo-looking myc.

So is powder agar, potato flakes, water and a drop of karo considered a high nute recipe?

Depends on how much you use. The ratios I have given are perhaps a touch high, maybe 3% nutes. I did that because its good for spore germination.
