I looked up whether DNA would degrade at 160 before I switched to 160 from drying at 125, and I found this study that found that "DNA in specimens dried at high temperature (up to 71°C) was not significantly damaged".
yes. i do have a reference... i dried Ps. cubensis in the oven at 120 one time...ate about 5 grams and did not "trip." you can either choose to believe me or not...
also...i'm not saying the DNA will be destroyed and unattainable. it will just be a lot more work and expense for whomever is doing the extraction and/or sequencing... and, not a lot of people have that to throw around.
I'm gonna choose not to believe you on that. I tripped pretty hard on an 8th of ovoids dried at 160 for 8 hours in my dehydrator and then boiled at around 212 for 15 minutes to make tea. A bunch of other people who I gave those ovoids out to said that they were the best mushrooms they'd ever had. I realize that this is just anecdotal evidence against anecdotal evidence, so I doubt this will do much to convince you. If psilocybin completely degraded at 120 like you seem to suggest, how would space brownies or any other mushroom edible work though? Also, I don't mean to be a dick, but can you cool it on the ellipses? I don't know if it's intentional, but writing "..." after everything makes your posts seem really condescending to me. Quote:

Are you referring to where they stated that "Some species of mushrooms (Grifola and Hypsizygus) showed slightly weaker PCR band when specimens were dried at 52 or 71°C (Fig. 4). ... In this study, we have not prepared replication of each species and we cannot conclude whether the infrequent presence of weak PCR bands is truly due to high drying temperatures. We argue it is probably not the reason because the same species dried at the the highest temperature (71°C) in this study showed strong bands (Fig. 4)." The lack of replication definitely makes this a flawed study, but this doesn't convince me that 160 does all that much to the DNA though. I just found this paper though, and it doesn't look so good for 160. I'm definitely switching to 105 for future finds. I went out Thursday to check for more at my spot, but I didn't find any Panaeolus species there. The season might be over, or maybe I just need to wait for heavier rains. Until I find more, which might not happen again until next year or even the year after that since I have plans that would put me out of state next summer, whoever I send samples off to for DNA analysis is just gonna have to deal with the samples being dried at 160. Sorry guys.Quote:
RuralAnomaly said: then, pray tell, how would one brew tea, with tepid water? (and i do know F from C) my take-away reading that was that getting the water out w/in 4hrs was primary, and then keeping the bits in the freezer afterwards kept things kosher, moreso than temperature degrading dna while drying. but it was a quick read.
i happened on a couple similar looking ones in june too - in grass - might be widespread in ohio? it'll be interesting to see what i get under a microscope too.
I think Copelandia spp. are probably pretty widespread. Just overlooked. It'd be cool to see if the mushrooms you found are the same species as mine. Definitely check the spot you found them at after heavy rains.
I might be bioassaying some of these today. Maybe I'll add a trip report.

I don't want to annoy anybody by bumping this but I received a print from this collection a while back....
I swiped spore from it on agar, cleaned the culture and inoculated 4 barely grain jars which colonized very quickly.
After spawning to hpoo, coir, verm, finely chopped straw, and gypsum, they colonized the trays in 10 days. I cased the trays with 50/50 and introduced into fruiting conditions today. One tray has a 1/4 inch layer of casing, one had just a sprinkling.
If anything happens I'll post pictures. The myc is whispy (typical of the Panaeolus genus) and a light bluish color. It did form blue sclerotia in culture.

Wow, how did I miss this thread...
I also believe they are Bisporus. Before reading all the microscopy comments
See stonesuns post on the Bisporus Collection I found in Eastern Wa, almost Identical to yours, dark thin caps heavy blueing ext.
