Question about Isolation

So I recently had a moderately successful grow from which I took a number of clones. Most of these clones developed multiple sectors (8-12) however one seems to be only 6 or 7 with one clear sector spanning about 160Β°.
My question is although I will still pull that sector to another plate, is it likely that this sector is a single strain?
I have 4 sterilized rye ziplocs I plan to use to test whole clone cultures, but if that sector is likely a single strain I will use one of them to test it and get a jump start on my isolate testing.

Ok... front and back, as clear as I can get them. (Overgrown... I know. Fuck real life responsibilities)
Bear in mind that I am isolating further on agar and will find out whether it is an isolate or not before it gets to fruiting, so a definitive answer is not what I am looking for. I am just curious as to whether it is likely or not a single sector so that it can be colonizing while I further isolate the strains from the clone.

looks like the whole top 180ΒΊ of the dish is your best bet to get an isolate

Right on. I figure on isolating the other strains too, mostly for practice, but that is the spot I was curious about... now to see how it does.

Personally I wouldn't even worry about getting an isolate out of a clone. Some people even think that its possible all the gentics are expressed in the different sectors anyway. Others suggest that intentionally stripping genetics out that originally fruited in concert may have drawbacks in performance. I won't say anything definitive but so far my own experience has lead me to believe that they might be on to something.
I simply look to continue with strong healthy growth. Avoid weak looking growth but I personally wouldn't sweat multiple transfers myself.

I certainly follow you on that Pasty. I was just going to isolate the clone sectors for the practice. After the transfers, the plate is getting diced and dropped in to jars anyway.
One quick, slightly off-topic, question while I've got you... I ran out of petris and figured on giving your plates a go but I have to use polyfill instead of micropore. Is the paper towel used to protect from condensation inside the pc or to soak up excess rh from inside the pp5?

Its optional. You don't need to have it and polyfil should work fine on its own.
