OP, I am a newb in this world...but that larger thicker looking spot and the one next it look like prime candidates for transfer. Everything to the right and top in the photo in the liquid looks contaminated to me.

I would wait another day or two and then transfer from one of the two big blobs in the center. Stop peaking into the petri to take pictures

I have just done 6 transfers from the original plate. Waiting time now for them to get going.
What do I do then?

Isolate further to find a monoculture if you want (might take awhile) or put a wedge on grains and grow it out. After that, restart the process by cloning the best performing mushroom, and isolate again!

If this is your second or third transfer I'd start thinking about what grains I wanted to use

I really hate popcorn, I think it's huge grains that take up a lot of space, look like shit colonized and tend to pop while hydrating or in the PC.
Plenty of great WBS teks: http://www.shroomery.org/forums/showflat.php/Number/17252080/fpart/all/vc/1
The point of spawn is that each individual kernel becomes an inoculation point in a bulk substrate. In theory (And practice ) the smaller the kernel size the quicker it would colonize a tub. If I had a jar full of marbles and a jar full of a brick the brick would have to expand and slowly colonize everything while the marbles would just expand and meet each other.
With that long-winded response out of the way, go WBS

What am I looking for in a good monoculture?

Good fruiting potential, fast colonizer, great pinsets, 2-3 flushes. Monos are great, it's just tough to isolate and isolate without knowing fruiting potential IMO.

Also, you can try oats, it's cheap, simple, and there's a bunch of teks out there.
