That is heavy with bacteria. The first pic is almost all bacteria that's been colonized. You can tell by the extremely thick white cottage cheese look all over except that tiny clean spot . The "piss" (proper term is metabolites, other members get pretty mad if you call it piss. Metabolites help fight against competitors. They also help "metabolize" and digest a substrate) is its reaction to the bacteria, and its response to fight it.

tl;dr it's pretty much done. I would not trust that as spawn.

What are your Sterilization and inoculation techniques?

Thanks for the response, i am still learning and researching.  I use rye berries.  For sterilization, i use a small bathroom with no windows, clean it up really good and bleach it down.  I use syringes to inoculate and i my my own homemade incubator that keeps them around 82 deg.

Bathrooms are extremely dirty man , no matter how well you think you are cleaning.  Also you don't need the incubator.  Bacteria thrives in warm temperatures so you're giving it a head start so to speak.  How are you sterilizing your grain?  How hot and for how long?

Some are, some aren't. You just don't want airborne contaminants flying around. I did my sterile work in a bathroom for a long time, no issues there.

same here(mine is very very clean), I kinda turned it into a still air room. worked great every time, even did agar work with a ridiculous success rate.

Ok. Well do you use SHIPs and a filter on your lids? Even stuffed poly is better than no filter. You should also think about making a sab. I can guarantee a still air box(sab) in your still clean bathroom would work BEAUTIFULLY and you could even do open air agar work in it. Check out the improve your sterile techniques link in my signature and read it top to bottom if you're serious about being successful 99% of the time. It also has a way to make the sab in like under 5 minutes. You just need an empty coffee can, a 100+ qt clear tote and your stove top

Also how much solution do you do per jar? With pf tek you might do 1 cc per jar, but with grains only do 3 drops MAXIMUM down 1 side of the jar. Let it grow until 20-30% colonized, then shake it to distribute the grains. You can't shake pf tek. That's why you have to use so much solution on all 4 sides.

Using less solution makes it so there's a much lower chance of other contams in the syringe to come in. Also 1 drop will still have 500000-5 million spores in it. It doesn't matter if there's 1 million or 20 million spores. Germination rates will still be the same. Hope this helps

I can tell you all my agar work ever only uses 1 drop, and it gets distributed A LOT in my dish.

Yes I use a still air box i made with plexi glass and cardboard box and inside is my tyvex suit i use in my bathroom in which i clean the shit out of with bleach and lysol. I use an alcohol lamp after every hole.  About the CC i used 2 per JAR!!! that might be my problem!! i was successful doing brf tek and i did 1 cc per side was not aware only to use drops on rye grain in QT jars.  Must have been using way too much and i will def take a look at ur sterile technique, i really want to get theses going, but the science behind these intrigues the hell out of me.  so interesting!!! thanks again guys!!

:rockon: also I forgot to mention that you don't need to incubate during colonization and fruiting. Colonization and fruiting at 65-75 will be much better. Anything above 80 also promotes bacteria. I personally colonize at 65-70 normally.
