Storing petris on their side so myc grows upwards & bacteria stays low down
    #22292122 - 09/26/15 07:52 AM (8 years, 4 months ago)

I read this old thread
http://www.shroomery.org/forums/showflat.php/Number/18569849

where a guy uses jars as petris and coats the sides and bottom. Transfers to the side and turns the jar upside down.

What happens is that the desired mycelium grows upwards over the peroxidated agar film (in the direction of the peroxidated agar layer) while contaminants, if present do not only need to fight the peroxide but if they are bacteria they tend to drop downwards (in the direction of the lid).

Stephen L Peele had a technique where you would pour hot/warm agar on top of colonising agar and the myc would grow up through the new layer. Also said to leave the bacteria below and you then transfer the newly emerging myc. RR used this too.

I currently have a pin transferred to a small plate. There was myc and what looks like bacteria growing. I put the plate in a small zip lock back and have turned it on its side, the bag stops the lid falling off and keeps it from rolling around. You could just wrap it in film but you also want to stop it rolling like wheel.

It does seem like the bacterial looking growth is dropping downwards in the plate and not growing up. Also it appears to be pooling down the bottom, like new liquidy growth is rolling downwards which should help it stop spreading so easily.
