Cloning from a dry mushroom?

Hey everybody thanks for giving this a look. i was just wondering if someone here can answer some questions about cloning a dry mushroom and the cloning process. Basically my questions are is 1) what is the best area to cut from a dry mushroom in order to effectively clone it on agar? and 2) From my understanding when cloning a mushroom instead of getting spore prints and using spores to inoculate jars, instead you get mycelium growth on the agar plate. How can I, or is there a way to use this mycelium in the standard BRF PF Tek? Thanks guys

let it go more than 19 days so it's fully dormant and dried. Try it with tissue a year or more old. Normally, it will take two to three weeks or more for mushroom mycelium to begin to grow from totally dry tissue. Often, growth starts sooner than that, but is almost always mold. If you have growth from completely dry tissue within a day and a half, it's almost a guarantee of mold. You can quickly determine that under a small microscope. Mushroom mycelium will be straight, while most molds will have an appearance of a steel wool pad with kinky, twisted mycelial strands with lots of side branching. Good luck.
Iodine is a great bactericide. I dip dry tissue first in iodine for a minute or two, then wash it off with a peroxide dip for five full minutes, then take the tissue through two distilled water washes before placing on the antibiotic agar. This procedure works for about a 10% success rate. Usually one or two of the cultures will grow mold, one or two will grow bacteria, one will grow mushroom mycelium, and the rest just sit there doing nothing. One of the keys for successful dry tissue recovery is to inoculate as many petri dishes as you can afford in order to use the numbers to your advantage. RR
I have never tried but RR has
You want to clone from the inside of the stem. BRF paste and whole rice have been used in place of agar but I wouldn't trust it with such a fragile project.
You can do PF tek from LC or GLC made from a clone. Make sure to clean the clone up and isolate all contams before doing any type of LC though.

Thickest part of the inside of the stem. Also try some gills. Mine had some parts of unbroken veil as soon in my avatar. I removed the unbroken veil and pulled out a couple of stems. I am also wondering how to transfer from afar wedges to brf cakes and learned the you don't really need the dry layer of verm. Or any other filters. People said it works fine with only to cover the lid with foil.

Damn im hoping the mushrooms i have will do they're pretty small the caps are about a half inch to 3/4 inches with hardly any stems anymore, and the stems i do have left are mostly hollow. But its worth a shot anyways

i think theyll work though theres a few that have the caps still completely untouched and attached to the stem with veils so id assume it should be as sterile as they'll get
