UPDATE!! Agar Plates Have Growth!! Finally made it to grains!

Many of my first attempt agar plates with Argentina MS have growth! Only two have no growth and 1 have a contaminant, but looks like I may still be able to isolate the mycelium out? What do you guys think? How long do I let the first round go? I was under the impression with MS you shouldn't let it go too long before transferring to a new plate.

I usually wait until I can divide a couple of good looking spots like u have there into three wedges and from m there make 6-12 new plates one or more will be pretty close to a mono then.with one.or.two more transfers I get my mono then just make how ever.many transfers I need from that. Just an idea.

Also, do I have to cut a wedge or could I use my inoculation loop to pick up a portion for transfer?

Get a scalpel or an exacto knife, flame sterilize until red hot then cool it in the receiving dish. Cut out a small square, scoop it up and place it in the center of receiving plate. Wouldn't make much of a difference to put in the fridge if you are transferring soon

I agree , from the germination plate I take a transfer as soon as I can see clean, healthy growth. I take it when its grown out a little less than the size of a dime.
Next plate I let grow out a little further so I can try to pick out aggressive growth.
Third plate is usually good enough to put to grain

It really doesn't matter :smile:. The entire wedge will get fluffy in 24-48 hours post transfer

That wasn't nearly as hard as I thought it was going to be in my SAB. I will need to bring home some parafilm from work because working with saranwrap is ludicrous.

So of the 13 plates I had ready to go, 2 looked like they could possibly have contaminants brewing and 1 had too much agar so I threw them out. I transfer a total of 10 wedges from the 4 plates I had. One of the wedges flipped while I was wrapping the dish and rolled around on the agar before rolling back into position. I wonder how badly that plate is fucked up?

ooks good man! Wrap them up better though.. cut a 1.5 " section off a roll of cling wrap if you don't have paradigm so you can wrap the edges and pull it tight

I brought some parafilm, but I figured I would use it after next transfer. I now it doesn't look like it, but this is saran wrap. The crap wouldn't stick to itself which seems ridiculous. But it was 2.5 inches cut off the end of a tube. It seems pretty tight, but I added tape at one end to hold it in place. Should I leave what is in place and add another layer?

Oh wow that is beautiful! LOL! I pulled that crap so tight that I accidentally flipped one of the wedges. Maybe I need to cut less off the end of a tub or just use the Parafilm. Although the parafilm won't like being over saran wrap.

All of my 1st wedge transfers look like the photos below. Out of the 9 remaining plates, 1 was contaminated. Time to transfer again?

yes, transfer again! 

plate 1:  transfer from 2:00
plate 2: transfer from.... uhhhh. maybe 11:00 or 1:00 :shrug:
plate 2: transfer from 10:00

you could also do multiple transfers per plate

good job :thumbup:

You should really buy 4. I bought two and as I am PC'sing my agar now I am completely regretting that I didn't buy 2 more. They are fricking awesome!! A little spendy, but totally worth it. I got the 500 ml bottles instead of the liter because I have small hands and wanted something small to handle in my SAB. I used a damn 750ml sake bottle last time and I spent more time worrying that I would drop the bottle that I overfilled some of my plates.

And thanks for the encouragement. This stuff is addicting! I am so close to spawning my first jars to bulk this weekend! I haven't even considered what I am going to do with all these mushrooms if I am successful.

Having a ton of Mushies is a GREAT problem to have...lol
They keep pretty well if you dehydrate them to "cracker dry" with a food dehydrator... I highly recommend a dehydrator if you don't already have one! Keep up the GOOD WORK, You are Rockin' It:rockman:

Hahaha I haven't even gotten there mentally, but at this rate with 24 quart jars and agar in the works with 2 syringes remaining, I am going to have more than I had ever imagined. It's somewhat terrifying how quickly this stuff accumulates and how I just keep trucking along. I guess I am starting to know what it feels like to be a hoarder or a crazy cat lady...

Yea it takes a little practice. I wish I could post a video cause there is kind of a trick to it

I think you're right, maybe cut the roll just a bit shorter and it'll make it easier to manage. That is the worst when you knock the wedge around and cant get it back in place, it leaves myc all over the place too when your platr grows out .
You wil LOVE parafilm though!


You are doing a nice job with your agar work, keep it up!!

Transferred a shit load of wedges to a plates poured 48 hours ago. The process keeps getting easier and easier as I go. This time around I tried two different recipes PDA and MEA... first two rounds were on MEA, but I felt like experimenting with different types of agar to see which recipe the mycelium liked more.

My plates are labeled and divided based on 3 things: Standard transfer, fuck up transfer (rolled around on agar or the petri dsh had more condensation than I liked), and contaminated transfer. Luckily I only had 2 contaminated plates from my last batch of 11. I didn't identify which were PDA or MEA because it is easy to identify by color alone.



So much fun.

Its not.my idea exactly i just added a little to it. I used a soldering iron to melt.the edges of the discs.

Here we are: Day 2 after transfer #2 and things are looking good. None of the plates are showing signs of contaminants. 3 of the PDA plates have condensation and the wedges flipped off to the side on 2 other plates, but I would say things are looking great!

Day 4 after transfer 2- 3 plates have green contam. Otherwise, seemingly looking good. Give this is my 3rd plate- Do I let this one ride out a little more? MS>Wedge Transfer>Wedge Transfer

OMG what the hell am I going to do with all of these plates? I sure as hell am not going to spawn them all at once. Plus I already have 2 tubs already in colonization, and 12 jars of oats almost done colonizing. Hahaha. I think I may have bitten off way more than I can chew, let alone swallow.

Haha. Yeah i did the same. Endend up chunking alot of petris myself. Made some lc and put it in the frige. But yes normally if i do like 12 dishes with ms. Then by the first sometime or even the second ill have a mono. But by then ill have almost 35 dishes. But i wont have to transfer as many times and its alot faster for me to get enough monos to start knocking stuff up. Its a waste for sure but it takes half the time.

If you have excess colonized clean plates you can stick them in the fridge to store and drop pieces to grain later. Clean out/ kill the dirty plates and you can add more agar later (I'm not sure if this works for disposable plastic plates, the PC may melt them)

Actually that seems to make even better since than ironing the whole material as that seems to prolly allow the pollyfil to breath still well, but at the same time maintaining consistent shape!

Lol yeah shes a beut! And yea once u get a good mono culture do some slants. I wouldnt slant an unclean culture imo though. Plus once u decide to use a slant ur gonna wanna put a peace on agar again to confirm its still clean to be on the safe side. And to make enough to innoc. Better to add something clean too it first ya know? Save u some worry.

UPDATE!! So stoked to have just completed my first cycle with agar. Today I transferred a dime sized wedge from 6 plates to 6 jars of oats! Check it out! I couldn't have done any of this without all of your support and advice guys. Thanks.

Not sure on the time line but you can do that or you can save a small sample of the Agar and transfer it to another plate and keep it in the fridge. If you like it you can then take that and put it on a master slant. The grain jar is also an option but it seems like a waste of a jar to me just my 2¢

Congrats on making it to grain!  Not to thread-jack, but I'm also having some success with Agar and culture isolation. 

I had 2 decently colonized plates, so I took a section from each (2 from each plate) and put them in their own on 10/10. 



Growing out nicely (though small).  Seven below isn't contammed, it's just got a scar from where I, like a dumbass, cooled my knife in the center of the plate, any black specs you see are char from said knife.



8 broke into a bunch of pieces so I just left 'em where they were.  Even that's growing well.  I'm most hopeful about 9... it looks very clean with some nice growth as far as I can tell.

Sometime's it's good to store a culture that isn't been isolated yet so that you can still go back in case you isolated something shitty.

Thanks Inoc. I really enjoy working with agar. I seemed to have more luck (more aggressive growth) with MEA over PDA. However, I was using PDA which was probably formulated for bacteria over mycelium. While the PDA plates were clearer and easier to look thru, the Mycelium seemed to really like the MEA more. What's your preferred medium?

I mostly use Pasty's PDA recipe, but I'll switch between that and grain water agar for some variation in nutrients.

I had to google Doxing. I figure if anyone is gathering information on me, they will find something whether it's there or not. But I'm a tax paying, mostly law abiding citizen cultivating for hobby purposes. People are funny. I can't believe I'm about to say this, but I'm in a holding pattern as we speak and don't have a single thing to work on until some of my current projects come to fruition. Agar transferred to 6 quart jars of oats. 2 tubs put into fruiting conditions, 2 tubs colonizing, 4 jars MS to oats same strain (not portobello, oysters, or shitaki) as the agar wedges I transferred. 10 jars of B+ MS to oats and 20 plates of B+ on MEA as we speak.

I feel very productive considering this is my first go at any of this and part of me wouldn't be all that pissed off if I failed miserably. I'm just loving the process. But I can't help but hope that one day my dreams will come true (yes I have dreams about the whole process)... To see pins, then immature fruit, then the final fruit of my labor.

If you're in a holding pattern, go buy more jars or learn as much as you can.  If you're all mushed out, learn about something scientific like the taxonomy of life or something.

And yea I think we all have dreams about growing...  the first time I tripped on my own mushies, I was walking on a gravel road.  When I looked down, instead of rocks, I saw little pins growing before my eyes.

or... just ask your bf/husband to jerk off into your substrate

Lol. I just figured out how to click on blacked out text to read it. I nearly spit out my coffee.

I keep myself busy. No worries there. I was starting to worry that I might end up buried in pounds of mushrooms. I got caught up in the zone of cultivation until my boyfriend looked at the tubs and said, " I think I might be too old too enjoy this trip." What that basically means is, "Whoa buddy, have you thought about what you are going to do with the end product... Because you might just be enjoying them alone."

I enjoy solitude. I live in an area that has excellent outdoor areas and I have a weekday off from work! But... I should take a minute to consider how much one person could possibly need. Waste not, want not.

I'm still eating mushrooms from my first monotub, I got about 15 more tubs that didnt contam out done since then...they just keep piling up and I don't mind.

That may be correct. I like growing them more than I like eating them. If I do eat them, I'd rather eat them fresh, they taste a whole lot better that way.
