UPDATE!! Agar Plates Have Growth!! Finally made it to grains!

Many of my first attempt agar plates with Argentina MS have growth! Only two have no growth and 1 have a contaminant, but looks like I may still be able to isolate the mycelium out? What do you guys think? How long do I let the first round go? I was under the impression with MS you shouldn't let it go too long before transferring to a new plate.

I usually wait until I can divide a couple of good looking spots like u have there into three wedges and from m there make 6-12 new plates one or more will be pretty close to a mono then.with one.or.two more transfers I get my mono then just make how ever.many transfers I need from that. Just an idea.

Also, do I have to cut a wedge or could I use my inoculation loop to pick up a portion for transfer?

Get a scalpel or an exacto knife, flame sterilize until red hot then cool it in the receiving dish. Cut out a small square, scoop it up and place it in the center of receiving plate. Wouldn't make much of a difference to put in the fridge if you are transferring soon

True. Wouldn't hurt, may slow them down until you transfer.

I agree , from the germination plate I take a transfer as soon as I can see clean, healthy growth. I take it when its grown out a little less than the size of a dime.
Next plate I let grow out a little further so I can try to pick out aggressive growth.
Third plate is usually good enough to put to grain

I'm about to transfer. Do I place wedge face up or face down on agar? I had a link once with pictures of the process, but I can't find it. Can someone help me out?

It really doesn't matter :smile:. The entire wedge will get fluffy in 24-48 hours post transfer

That wasn't nearly as hard as I thought it was going to be in my SAB. I will need to bring home some parafilm from work because working with saranwrap is ludicrous.

So of the 13 plates I had ready to go, 2 looked like they could possibly have contaminants brewing and 1 had too much agar so I threw them out. I transfer a total of 10 wedges from the 4 plates I had. One of the wedges flipped while I was wrapping the dish and rolled around on the agar before rolling back into position. I wonder how badly that plate is fucked up?

ooks good man! Wrap them up better though.. cut a 1.5 " section off a roll of cling wrap if you don't have paradigm so you can wrap the edges and pull it tight


Ok maybe this is a bit sloppy too haha but it's the only pic I have around..

I brought some parafilm, but I figured I would use it after next transfer. I now it doesn't look like it, but this is saran wrap. The crap wouldn't stick to itself which seems ridiculous. But it was 2.5 inches cut off the end of a tube. It seems pretty tight, but I added tape at one end to hold it in place. Should I leave what is in place and add another layer?
