hey guys!! so i won a red boy print from stropharis a little while ago. so i made some of my first agar dishes (pastywhites no pour) made sure my ratios were right. that it was the right consistancy. then made some more and transfered some spores to the plate using a sterilized (in pc) qtip, on 9/14. today i finally see growth.
so my first mistake was using quilted 1/4 pints. i wanna use the 1/4s cuz i can re-use em for mini cakes. so yesterday i went and got some more 1/4 pints (the unquilted kind) and some pp5 mini-rounds and made up little more agar.
what i wanna know is whether its time for a transfer. its hard to tell with the quilting. i couldnt get you a pic without removing the lid. i placed the spores a little off center, but the white growth is directly in the middle just off the side of the spore load, and fuzzy, not rhizo. i also see a couple white spots in other places away from the spore load. so im a little worried. although spores could have fallen off the qtip and floated to other areas i suppose..
so what do you think. at least do a transfer to get it to a more visible container(and pics for you since ill be taking the lid off)?

Do you have a SAB? Open it up in the SAB & take pictures.

Yea those quilted side jars suck sometimes when you're trying to distinguish what's going on. I usually will let my transfers grow until they're a little larger than a quarter, or unless I see a contam I can get away from.

 i cant get the camera to focus on the jar very well. it tries to focus on the SAB. so frustrating..

Yea man take a transfer. On first plate you want to take a transfer as soon as you see growth big enough to take one really, I usually do when growth gets around the size of a dime tops.
Don't worry if its fuzzy or rhizo, just take something that looks healthy to your eyes. THe first plate from spores is usually really fuzzed out and hard to tell whats what anyways so just take a piece that looks healthy and clean and is furthest away from any bacteria or mold colonies.

what are your favorite colors for agar? iv tried this blue and a purple that turned out more pink. i like blue more.

Ive never used color in my agar actually so I like to get mine as clear as I can. But I might start using it to distinguish between different recipies/antibiotic agar and what not

well i dont think it matters much but heres an update on the first dish after it grew out some. kept it to check up on my sterile procedure after the first transfer. i dont see any contam. i kno its a little blurry. but i think i did okay. how long would it take for them to show if my procedure contaminated it?

No way to know really, just let the second plate grow out some... like the size of a nickel or a quarter (unless you see it growing towards bacteria or mold.. then take a transfer asap).. and try to take another transfer from the leading edge of the healthiest section

just wanna say iv used agar for all of 6-7 days and i already love the stuff. i had an lc that i did some noccing with and they all came out bad.(b4 i had the agar) so 2 days ago i put a little squirt on some agar to see what was goin on. apparently ALL the white growth suspended in that lc is trich. not a lick of healthy nada.
so yea agar ftw buuuuuudy.

i know i cant wait!!! especially when these boogers show up in the mail.
 
but on to a little more on track discussion. heres an update of transfer 1 after 4 days of growth.

update: the growth took off a bit from yesterday and is about the size of a nickel. one side has definetly grown a little faster/farther. how long to let it go before should i take another transfer?
btw id like to make and test some isolates to find a good fruiter. thats what this thread is planned to develope into. but until then id just like to have something clean to work with.

update: the red boy slice is now totally fuzzy and jumping on to the agar. id say a couple more days and itll be ready.
in addition to that i recieved a syringe of pan cyan that i put to agar as well. my problem is that the mycelium thats popping up has a bald spot in the middle of the germination points. normal or no?

edit: this is my first pan cyan grow. the tissue around it looks like the fluffy white mycelium i see in pics. just the bald spot seems weird..

so my RB plate is starting to take off again. its not showing any strong rhizo growth but its plenty clean now. i dont see any contams and iv been checking all the plates iv transfered from and no contams. my original T0 dish is actually totally colonized.
so what would you say my next step should be. continue transfers till strong rhizo growth?
or straight to tiger drop plates?
i intend to get a good clone culture to work with. so im not sure if i want to limit genetics much more. advice?

i have been very lax on my updates but progress on RB has been slow which was surprising. seemed as though it was much faster in the beginning. but the plates i used were a couple weeks old so maybe they were dry? i dont know. but the RB-t3 didnt look to good. it didnt look like contams but the mycelium was very fluffy and airy most of the way around. i found one good rhizo sector and took a xfer of it to some newly cooked plates.
my pans however seem to be doing just fine on the plates. took another strong sector and transfered to t3
