4 Days Post First WBS Inoculation

I've been a good girl and despite the urge to look, I have kept the dozen quart jars I inoculated with a Golden Teacher syringe on 9/15 out of sight.... until this evening. My home has been kept at a comfy 75 degrees F for the last few days and the jars have been in a cabinet where the door doesn't close and ambient light makes it way during the day.
I was thrilled to see that all but one lonely jar are showing signs of growth along the glass just below my silicone injection ports.
So, should I look daily now or wait another 4 to 7 days? Do I shake when the exposed grain looks about 40% affected?
Ha! I'm so excited!
In the meantime, I am knocking up some agar tonight. Thanks for the advice guys. I couldn't have gotten this far without you.

Look at them as much as you want... the less you do the faster they will seem to grow 
Yes, it's good to shake around 1/4 - 1/3 colonization to help speed up the process.

I'm a very patient person.
I think I may have overfilled my jars, so I think they may take a little longer than I planned to complete. I filled them just over half way before throwing them in the PC. It seems that they are about 3/4 of the way full now, but by no means does it look like there is too much moisture or they are too closely packed.
I'm curious how long after initial growth I should expect until I have to shake?

It's always a roll of the dice but I'd say within 1-2 weeks from now you'll be good to shake.
You're right, you do have too much grain in your jars. It will be ok, just a pain in the ass to break apart.

I used gypsum and finch seed without sunflower seeds or corn. I moved pretty well after PC. I'm hoping it's not too much work but at least I'll know better for next time.

I can't believe how much mycelium developed overnight! All 12 jars have growth and no signs of contamination as far as I can tell. I am curious if it is normal for there to be a trail or birdseed and mycelium from my inoculation point. I intentionally did spray the 0.75ml along the side of the jar, but I didn't expect for the mycelium to grow on glass like this.
I know my jars are overfilled, but they look even more overfilled in these photos due to the seed and mycelium along the glass. I took a photo from the other side so you could see that the jar isn't completely filled.




Petri dishes were knocked up last night. No growth yet.

It's growing where the spores dripped. That is normal.
From your last pic your jars don't look over filled if they are all like that.

Yeah, all but 2 are exactly like that. The other 2 have more but not by much. So far all have great growth but one... The one I failed to shoot the solution along the side of the jar. I squirted it straight down and it looks like the mycelium is growing on top of the seed in the jar and at the bottom of the jar a little. I suspect there is quite a bit hidden.
48 hours since I inoculated my agar. No growth. No contaminants.

You used MS I'm guessing? How many drops did you put on each plate? It'll take a bit longer for the spores to germinate. Be patient, it'll grow.

Caelistis, Shake those jars. Mix up all that stuff. Break up the streaks of mycelium. Yes it is totally normal to see it like it is. I prefer to squirt the spores down the side of the glass. That way I can watch the growth and look for contams. But now shake them, and try to get all the grain to settle towards the bottom. So tap the bottom a few times after shaking.
The agar could take a few weeks.
-------------------- Stop experimenting half way through your first grow. Grow it to maturity, watch it, learn from it. Do this a few times then experiment with different ideas and figure out what works best for you.

I used as drop of MS on my inoculation loop and against my better judgement but at the advice of the forum scratched into the agar with the loop. Definitely not how I'm used to doing my plates but this is a new realm for me. I did a few just by adding a single drop with needle and a few with the loop not scratching into the surface as well. I have them incubating at 80 degrees. Is that too warm?

Thanks for this advice! I was going to wait until they were at 30% but I had a feeling that shaking now would work out nicely. I'll shake right now and keep you guys posted.

I would wait a few days til they look more like this to shake 
You only want to shake once during the colonization process, and if you shake now it won't be as fast as if you wait a few days. There isn't even any rhizomorphic mycelium yet.. its still just taking hold and will take longer to recover versus if you wait to see rhizos.

Well, I shook them despite conflicting advice. I'm glad I shook them though because the mycelium at the top of the jars along the glass was really stuck on there! I had to use a phone book to tap it off more so because I'm a weakling more than anything else. Hopefully I didn't ruin anything but there was about 2x as much growth as what I showed on the pictures. Now it just looks like seed with a coating on it.

False. Those jars In your pic are way past due for a first shaking. You can shake more than once. Depending on the amount of inoculate. But she can do a test and see. Shake all but two of your jars now. Shake those two next week along with giving the other jars a second shaking. See which way works better.

It's already done and I'm not too worried about it. I know that the hyphae are there and have the right conditions. I'll keep you guys posted!

My jars now look like they have stark white chicken pox after shaking. I'd say they recovered nicely!
