Agar to Lc

hi i was wondering how to best transfer my agar plate free from contamination to my Lc solution. i dont want to open the can to my lc for the contamination risk. any ideas?

You're gonna have to open it in an SAB and drop a wedge of the agar in there.

im trying my first agar right now. if im not mistaken the best way is to
equalize pressure in lc first
prepare your cutting
pop and slide the lid only a little
scrape the wedge onto rim of jar, making sure to keep your angle as verticle as possible to ensure contact only to the inside rim and not to where the metal band screws on.
then re-lid and shake.
you might could prepare your cutting with the dish on the lc jar. that way after taking the cutting, the motion of closing the agar dish can easily and swiftly turn to opening the lc jar. assuming ur using widemouth lc jar.. or i could be totally wrong and a tc might shoot me down.

I try not to touch my jar during transfer, especially dealing with water. I do what you said except take the ring off and slide the lid about halfway off.
I grab the wedge very loosely and it just slides off the scalpel when I hold it almost vertically over. I make sure the arm of the scalpel and my hand don't hover over the LC water.

In a still air box, etc. I cut the surface of the agar up. Just tear up the surface of a 1/4th inch square area or bigger if you want. Then scrape up the chunks of mycelium, and drop that into the lc. That way you are working with dozens of little shreds of mycelium versus a chunk. It will be usable in a few days doing it that way.

Why not just make a liquid innoculant instead? It's easier and faster. Why wait for your LC jar to recolonize, then have to suck up the individual strands of mycelium, that are sometimes hard to break up. Too much work IMO. I like to have LI syringes on hand and ready to go!

We have all had that dream of gallons of viable, clean liquid cultures. It's just not the case. I am never impressed with an lc after a month of using it. They get infected so easily.

I agree. I make liquid inoculate basically the same way. But instead of transferring the mound of shredded mycelium to a jar, I suck it into a syringe of sterile water.

check the videos in my signature for a no blender LI tek
im gonna remake the video soon, but you can also scrape the mycelium off the plate with a sterile syringe (inside a sab of course) if the power wash method in my vid doesn't work (it should).

I kind of explain how to do it in one of the post above. Instead of cutting an agar wedge. Take your sterile knife, etc, and scratch the shit out of the surface of the agar. Tearing the mycelium into dozens of little shreds. Now either scoop that up and dump it into your lc. Or take your syringe full of sterile water and squirt a few drops of water onto this pile of shredded mycelium. Then suck the wad back up. No need to cut a wedge and put it in a blender. Way too much work. Now you have a sterile syringe of liquid inoculate. Use a big syringe if you can. It's the same amount of work to make a 10cc syringe as it is to make a 100cc syringe. There will be more than enough mycelium floating around.
Putting a pile of shredded mycelium into an lc is better than a wedge also. Instead of One big chunk of mycelium, you will have hundreds of pieces floating around your lc on day one. You will be able to use the lc on day 3. You could probably use it on day one if you made sure to suck up a Pieces of mycelium.
