Inject agar & spores into disposable petris while still in their sleeve.

I had no SAB handy and had an unopened sleeve of disposable petris. I got a 60ml syringe full of sterilized agar and just injected it into the petris through the plastic sleeve.

I think they are put in the sleeves and irradiated to sterilize them. So inside the sleeve is sort of like a mini SAB.

I wiped the outside with alcohol but did not bother flaming between injections, which some might prefer to do.

I injected from the top down, but I think going upwards might be better. The petris are very light and there was enough room in the bag so I could use one hand to pinch the upper lid and crack it open slightly, then inject into the bag and squirt warm agar direct into it. Then move onto to the next one. You have to be careful that you do not tip the bag too much or it could cause the agar to go out the side of the petri. This is why I now think I should have started at the bottom, by going bottom first I can leave them lay flat after injection, and as I open the next petri lid only empty petris will be above it with no liquid agar to worry about spilling.

This also keeps them nicely together. You could run a line of tape over the holes if you wanted to keep them cleaner, or wrap film around it.

I am also thinking I could inject spores into the dishes while still in the sleeve, which could keep them easier to handle and stack. Keep an eye on them and remove any contaminated ones ASAP.

I was planning on trying to find a strain that would fruit invitro at lowish temps. This would allow me to very easily have loads of petris on the go at once, I could easily have 5 or 6 sleeves of 10 on the go with very easy handling.

I got 1 sleeve of 10 small petris for €1.93 including postage on ebay, this is a standard "buy it now" price, not some fluke auction I won.

That is a great idea... You should probably write a tek on it with pictures if you find that it comes out sterile.

I definitely like the idea of using a syringe. I'll probably do what you did, but you didn't mention how you got the sterile agar into the syringe... Did you PC the syringe with the agar liquid? It doesn't seem like it would work with pressure differentials when heating.

I might possibly try this by doing a no-pour tek in a pint or quart jar, and use a sterile syringe to suck up the agar after PC'ing through an injection port.

Would you mind going over your method?

That is the usual way I would do it. I usually just microwave it in a filtered jar. A large 60ml syringe reduces the amount of times you need to use the SHIP which increases risk of contams and shortens the lifespan of the SHIP. It also means you can make a large jar with lots of agar and let the remainder set afterwards. Then if you need it again you can just microwave it or gently steam it again until it is melted again, as it should hopefully still be contam free.

I had 60ml agar in the syringe and I plugged the syringe needle with some a piece of silicone rubber. I was hoping it would withstand the effects of the pressure cooker. When it was finished it had blown out and half the agar was gone from it so I would not bother trying again.

The other method I have used was having a syringe needle attached to a thin flexible silicone hose, this was then attached to a jar of agar and it was all sterilized together, there was a cap on the needle so agar would not flow out. My plan was to have a needle on a tube like on a petrol pump, where you could have the jar standing still and move the syringe needle about on a flexible tube and move it between plates easily within my small SAB.

This proved difficult, the agar would begin to set in the tubing and not flow, I had to massage it out. If it was hot enough to flow easily it would induce condensation and was harder to handle. The way to stop flow was to pinch the tube or move the needle upwards, this proved messy as you might imagine.

As I am not using a traditional SAB filling these sleeves I now have much more freedom of movement. My new plan is to get a jar and on the cap near one edge I will fix a syringe needle in place, on the opposite side I will have a filter. Now I will sterilize the jar with a cap on the needle. Now when it is still liquid and just warm enough to handle I will lift the jar and tilt it a little (no agar pouring out), then pierce the sleeve and direct the needle into the petris, now I tilt it more so agar pours out and tilt it back up to stop the flow. Air will come in the filter so it should freely flow out. I will practise this with a dummy setup first to see if it is feasible.

I may have to tilt the sleeve of petris to do this well. If I leave time between each pour the agar might set enough so it does not slosh about, the bulk thermal mass in the jar should keep it liquid throughout.

I have also considered making a giant syringe from a cake icing injector

or from a coffee aeropress

But I am not sure how well they would stand up to lengthy sterilization cycles.
