If any of WBS shows on glass, does jar have bacteria? Pics included.

I've had issues with bacteria in the past, but I started getting it right and I was using too much LC in my jars, which caused constant bacteria contamination.

I remember seeds would show on the glass without the mycelium covering it like the rest of WBS. Is this happening in these pics? I'm spawning tubs right now. I soaked these WBS for 30 min after a 12 hour soak and then I dry them on towels just until they are dry to the touch, but obviously hydrated and swollen with water (has worked in the past). I'm fearful that it's bacteria once again on the side of my jars.

The first 4 are the 1st jar, the last 4 are the 2nd jar. Both are being spawned into 1 tub.

It could possibly be stalling or the WBS is tight against the glass.

Except if it was stalling it wouldn't be colonized and if it was tight against the glass the texture would appear flat.  Good guess though.


It's either contaminated or it's fine.  Have you been kind of.. sliding it up and down the glass?  It looks like the mycelium was torn off of the sunflower seeds and that's the only way I know to do that.  How recently was the last time you shook it?  If you've only shaken it once, you can afford another shake to see if it recovers and that will tell you definitively.  The only reason I think it might be contaminated is because the way the mycelium looks is just slightly off.  Might be a small amount of bacteria present.  Not too much to spawn with, but probably more than you want to G2G.

Yeah, not sure why you suggested it was stalling, since it's 100& colonized...

Inocuole: Thanks for the advice. I shook it on the 11th, so just 5 days ago. It was a G2G from a jar was started by a clean agar clone. Shaking it may have pulled the mycelium off of the sunflower seeds, but my concern was more with the millet, how it is showing against the glass like that.

What did you meant the myc looks off? I've been at this 2 years and I would like to have an eye for bacteria, even though sometimes it doesn't show at all. Of course the WBS smelled fine, but that happened a lot when my tubs were contaminated constantly with bacteria.

When you say it's a small amount of bacteria and "not too much to spawn with," do you mean that it won't effect the sub? I would think ANY amount of bacteria would cause mold before the 1st flush. Obviously I woudln't do G2G with an iffy jar, so I'm just concerned now if it will fruit just fine for a few flushes or not.

I don't know how I could fuck up a jar that was started with a solid agar clone. I hope that the other 12 jars that were knocked up by this 1 agar jar aren't bacteria ridden as well. Do you think that all 12 would be contaminated if just one is contaminated?

Also, if I shake it now, will that small amount of bacteria really show up as mold and it will be obvious after a couple of days that it's contaminated? Most of my jars have these TINY little WBS spots where they're not covered in myc. Also, there's condensation on the glass where the seeds are, so some even look a little wet around the individual seed, but the inside of the jar is sweating either way. Is that bad or normal?

What even causes that wet look on the WBS? What mainly causes bacteria? I'm as sterile as can be in my SAB.

No, it just means that the grain is pressed up against the glass jar.  That's it.

I hope so. When I used to have such a bad time with bacteria, my seeds with have a noticeable ring of water around them on the glass, whereas these jars it's not as bad. I'm hoping everything goes alright.

Have you had a lot of jars with the same non-colonized side of WBS pressed against the glass?

would wait and see on that jar. extend your PC times and be double sure your grains water content is great if you're experiencing bacteria problems.

Cannot be bothered to find a link, but I remember one thread a dude transered same culture to two jars, one pc'ed for 60 and one for 120 and near full colonization the 60 minute cycle jar had far more grains pressed against the glass. Not saying that it's conclusive but not a bad idea to extend the pc time!

ps spawning with bacteria is common because we are all for the most part amateur with no super insight into if there is mold/bacteria growing with the myc on agar. I have a pretty good idea, but I'm not 100% 100% of the time. But I would say it effects the yield, for instance your substrate or jars might have metabolites from fighting infection. Just like when you get sick, your body feels tired because it's fighting some infection, you will have less energy to preform the myc will have less energy to fruit. Hope this metaphor makes sense. Ideally start from agar with an array of super clean looking growth is the best way to have maybe one be a knock out cleanliness.

Filthyknees: What is perfect grain water content? They are simmered for 30 minutes with or without a soak first. The 30 min simmer really gets the grains fat and full of water and then I dry them on towels so that the outside of the grain is completely dry and rolls right off of my hand. Would that be sufficient?

I am definitely PCing 120 min now. I have been doing 90 min for a very long time.

It's pretty frustrating to have such a small change make such a big difference b/c my last grow and the 2 before that did very well. I had only 1 bad sub and the other 8 went on to 3-4 flushes. I used only 90 min PC there as well as everything else I did the same way. It's a process I go through with these jars, etc... So, It's confusing to say the least when you don't change anything, yet your yield goes to shit because of bacteria you can't see or smell?

As far as the seeds pressed against the glass, does it always mean bacteria? I have only 1 jar that doesn't seem to have really any of those seeds pressed like that. I'm using this one as a temporary master to G2G my others. However, I do have a number of agar looking good.

One more thing, the jars that I used to G2G to the jars above in the pics, were all from clean agar plates.

And what do you mean super clean agar? Either there's mold or there isn't, right?

The closet they are in is at a steady 79-80f. Would that encourage bacterial growth? Or only if there is bacteria inside the jar in the first place?

Lastly, if any bacteria at all is in the jar, will the mycelium not colonize over it or will the tiniest bit of bacteria ruin a sub no matter what?

Sorry for the million questions, but it seems you have the answers that I've been needing for the better part of a year, so I'm trying soak up as much knowledge I can about bacteria as I can, as it literally ruined every single sub I made for over 10 months straight. I kept trying and kept failing, but I knew eventually I would fix the problem, even if by accident.

I had some jars in my last grow that looked like yours and it turned out fine... I'm still learning, but I just assumed that when there are uncolonized seeds, I didn't get those particular seeds properly hydrated, so they don't get totally colonized... Like I said I'm still figuring things out myself, but you will probably be fine... Just be sure to smell them before you spawn... You just want that great fresh Mushie smell...Anyway... The bulk substrate that we all use is pasteurized.  That means there is bacteria in the bulk substrate. Bacteria isn't always a bad thing . The few spots in your jars should be fine I wouldn't use them for g2g . Bacteria can hide really good on agar a little bit of gentamiacine (sp) in your agar will take care of it.

That's somewhat relieving.

So, I did a little test and circled numerous spots where the WBS was pressed against the glass. After a few hours, the spots were gone! They colonized over the WBS, so I'm content for the moment.

Granted, not EVERY grain was colonized, but it showed me a progression from last grow. Here's pics of the said pics...

I didn't think of the possibility of bacteria being in my agar. What's this stuff I need t put on my agar plates to fight bacteria?

And isn't the little wet spots a tell tale sign of bacteria?

It's a spray, yeah? Any certain way you apply the Gentamicin to the agar? Before or after inoculation?

Does bacteria on agar have a specific look to it? Is some of it only seen with a microscope.

As for this jar below, I'm going to use for G2G. It doesn't seem to have WBS pressed against the glass. If the WBS isn't pressed against the glass, is that the only way to tell if your grain has bad bacteria?

While mixing the agar, before PCing.

And if you use it with every plate you'll never get good at agar and you'll transfer contaminants without seeing them.  The antibiotic won't stop the bacteria from growing into the grain.  Best to use it for cultures you absolutely need to clean up or spore germination or clones, that kind of thing.

You can G2G it if you want, but be careful.  Grain can be pressed against glass without bacteria, and bacteria can exist without grain being pressed up against glass.  It changes the morphology of the surrounding mycelium and it looks slimy.  Sometimes it just causes everything to stall out.  If it recovers from a shake, it's good.

Is that an all around truth? That if I shake it while it's 100% colonized and it recovers just fine, then there is no bacteria? How could such a small amount of bacteria stop the entire jar from growing after 1 shake?

Is this how i can know 100% if a jar is clean, by shaking it once more before spawning? If so, that's great. I never knew this fantastic piece of info.

It wouldn't stop the whole jar recovering, but it would stop at least part of the jar recovering, or when it recovered, it wouldn't look quite right, and that's enough to tell you what's going on.
