Progress! First Inoculation of WBS Down - Now On to Agar...

I just finished inoculating my first batch of 12 one quart jars with MS which were filled just over half way with WBS. I decided to use just one of my 10 cc syringes for all 12 jars since there is a much higher chance of contamination with MS syringes to grain, this is my very first attempt at any of this, and I will be moving on to agar while I wait on these jars in the event they are a complete failure.
I both loved the silicone (ease of inoculation) and hated it (mess..mess..mess). I'm thinking this next time around I might make my own injection ports to pop into the drilled holes or buy pre-made ones. I also think I will use clear silicone next time. I didn't have a hard time finding the holes, I just don't like the way the crap looks.
I am fairly certain one of the jars is a failure as the Tvek square wasn't properly sealed and some grain came out after the PC/first shake. The rest seem to have a good seal, but I also didn't realize most people do a layer of Tvek on the top and bottom of the lid until after I finished my food PC load. I'm hoping the precautions I am taking with sterility make up for my noob mistakes.



Today I am moving on to agar. Wish me luck. I'll post pics of my poor excuse for a SAB later today.
Thank you to everyone that has contributed to this forum over the years. It has been an invaluable tool. Without it, I think I would be lost!

Good luck on the grow man! And for next time, It would be a lot easier to just crack the lids in your SAB and inject onto the grains.There's really no need for SHIP's. It's good to practice your sterile techniques now, since you plan to move onto agar.
I used to use tyvek when I started out too, but now I like to use SFD's, they're less prone to getting wet and causing contam problems, and can be sterilized hundreds of times without reducing the pore sizes of the filter, which would inhibit crucial GE.

Thanks for the tips! Out of curiosity, do you just make one ΒΌ inch hole in the top for the SFD then? And do you use micro-porous tape to fix it to the inside of the lid or do you fix it with silicone?
I just purchased some SFD's online and am patiently waiting to get them in the mail. I only used the Tyvek because once I received the polyfil I realized it was going to make a huge mess and be huge pain in the ass. I also don't have a whole lot of confidence in the polyfil's ability to block out contaminants.

Yes, I personally use a 3/8" hole for GE. Make sure to put the SFD on the top of the jar. Add silicone around the edge of the filter and press it down firmly. Let it cure overnight, then they're ready to be loaded up.
Great purchase, they will last you a long time! Make sure to make cut-outs of the filter, no need to use the whole thing, that could get pricey. I use a quarter to trace it out on the filter, and usually get about 7 mini SFD's

Well, you know, I may completely fail the first couple times I attempt any of this. I'm almost expecting it ESPECIALLY since everything has gone so smoothly so far.
I bought 24 more jars today at Target. I usually buy online, but I wanted jars on hand pronto. This hobby is getting expensive.

You're on the right track man, agar is super simple, clean growth will be easy to see, then you can transfer to your jars for some clean spawn!!
