LC to agar plate (no growth)
    #22240880 - 09/15/15 11:13 AM (8 years, 4 months ago)

I've had a vile of LC blue oyster in the fridge for about half a year.
Last week (Tuesday)I went ahead and pc'd some LC solution (use sfd and self healing ports on my lids).  I pc'd some syringes as well.

I sucked up the LC from the vile, squirted some into my LC solution and squirted a few drops onto a pasty plate I made back in March.

The LC solution has grown out and looks good (as far as the eye can tell I know).

There is no growth in the mini round I can see... not seeing any bacterial growth either though.

So...opinions?  weak myc? Old agar plates (do they go bad if kept wrapped in tinfoil in fridge for 6 months?) Not enough time?

Whoa, your agar is entirely too old. I can't speak for cultivation purposes, but in microbiology we use our plates within 2 weeks... honestly, best practice is pour within a week and use within the week even when stored in an optimal laboratory setting. So I would bet that 6 months in your fridge, covered in foil isn't going to cut it. Even if the agar isn't contaminated, the nutrients degrade. Time to mix up some new agar IMO.

yea..perhaps so.  I have some oat water to strain tonight so I'll give that a go with the agar.

Made a new batch up last night.  I'll mess with em tonight.

Question. Pussyfart says one drop of lc for agar.  Sooo I'm just confused by this because the white myc we see floating around in the water in the syringe may not exit the needle in that one drop or is there microscopic myc all in the water and in that one drop that comes out??

TIA

I have used LC many times before. You should shake the syringe well before using. Even if you don't get a large piece of mycelium out of the syringe, there is plenty of suspended mycelium in the liquid to start colonization.
