Yeah, figured you meant for isolating further. But fuck that, I don't have time to run a bunch of tubs of isolates to try and find the prettiest one (not yet at least).

Yeah in the long run I'd rather have done the shitty legwork on finding a really nice isolate and you are damn close there.

No. But out of a dozen nice sectors I would bet at least one of them would be killer. At that point you have a good performing culture that is also quite young. That will let you push the working culture for a few more expansions and a couple more months. I usually see a drop in performance with a clone after 6-8 months and a dozen or so P expansions. An isolate could probably go longer, meaning you go back to your masters less.

Yeah. I kind of knew the answer to my own question. I was asking because I've heard you rave, on a few occasions, about how well invitro clones do. If your taking from a slant and putting the culture on agar, from that point you could turn one plate into a dozen plates and those dozen plates could last you a good while especially if turning those plates into LI. With that being said, performance would not ever be an issue. So with all things being equal, which would you rather have? Invitro clone? or isolate?

Well the clone gives you a damn good indicator of how well it will perform. It still needs testing but you have some idea at least. It takes a lot less time too. So I have a lot more clones in my collection.

At the end of the day performance is performance. But stare is pretty damn close to isolates so I would push through to see what he has. Thats just me tho.

Sorry. I know I jumped into the middle of the conversation. Yes. I agree that because stare has really nice sectors on an ms plate, that he should isolate them. For me, I just wanted to know whether isolates have an advantage over invitro clones because I really don't want to focus my efforts on them types of projects unless it was worth the extra time and energy spent.

Is it absolutely imperative that an isolate is used when conducting an experiment or can one simply use a proven clone?

Maybe in a peer reviewed publication an isolate would be required.  But a stable and proven clone is good enough i would say for our purposes. Unless something was stripped out,  which would happen early, most clones will be very predictable.  I have a clone that will always put out a single fruit way ahead of the rest of the flush. Does it every time regardless of sub, fruiting chamber, depth etc. I would say something that predictable can be used to control the genetic variable.

Usually comes up at the very edge of the substrate. Does this in both bottles and monotubs, lined and unlined.

agar so soft that the transfer wedge melts into the culture dish.
the transfer wedge is red while the receiving culture dish is green.
This is using Pastywhytes no-pour recipe using 2 grams of agar instead of 2.5g.

make clones from the pins then knock up some millet. Maybe some WBS or popcorn if I feel up to it.

I'm really debating if I will put a few mono tubs in my closet in front of the grow tent.


or if I will buy some smaller tubs and put them under my bed. I will probably just get tubs for under my bed. There is plenty of room and air circulation.

I do have a question that I would like your thoughts on.
Do you think a half-gallon jar of millet would have a good chance of stalling out if I used an agar wedge?

I really want to just run a few half-gallon jars. Mostly because I'm lazy busy. :bigblunt:

I did half gallon jars of millet way back when I was first starting. Biggest issue was the top grains drying out. I suggest it would be better to colonize smaller masters like pints then generously G2G them to the half gallons.

However if your very confident in your millet prep you could do large wedges right to the half gallon. I would be prepared to shake twice tho to finish quicker, that a lot of surface area to cover.

ha;f gallons with tiger dropped pasty plates work well....my only beef is the amount of half gallons i can fit in my pc and they are useless for g2g

If I can fit 4 of them then I am only losing 2qts during the PC cycle but half the work when knocking up. I'm okay with that. :rofl2:

Do I need to up the PC time for running the half-gallon jars?
I normally run grain for 2 hours at around 16 - 17 PSI.
