The structure of the myc in question is very similar to pans but it's a lot darker than any pan myc I've seen before(which isn't THAT much) lol and it's stuck flat against the agar. No fluff to it at all and almost has a shine to it. Any tips on isolating away from a particularly persistent contam including those that slightly outrace the leading edge. I have a mazatec culture with some similar looking contam. It keep pinning ball shaped pins. Not sure if that because of the contam or if it's a mutation yet though

I guess we are talking about the Senguio? Well it's from jake so we know it's a pan.
I have some plates that look like that myc. It's from a wild print from South Africa. Not as bad as in your pic. I'm at T6 and still cleaning it up.
First pic looks like you can transfer from 9-11 o'clock This is the same plate from the previous post right?
And keep your transfer size down. Try to get a piece that's smaller than a grain of rice... right from the leading edge. That Senguio is only T2. I would expect many more transfers before that's cleaned up. And if it gets clean but slows down...you might as well throw it out and go back to spores. I've learned that lesson the hard way.

It does indeed. But as far as I can tell it does stop growing out. I'll put dishes in the fridge if I get stacked up on grows. So, I don't know that you could outgrow contams in this way.

Was my thoughts as well but was just a suggestion. I gues transferring and using antibacterial is the only way to go?

Sometimes it's best to go back to spores. Like goddamn I am having to do that now. I just threw an ass load of plates in the trash and am starting over. I've seen many times that you don't always get what you are looking for.

Thanks for the tips. Yes that is the same plate and yes it was from Jake so I know it's definitely what it's supposed to be. Thanks again Jake even if it's giving me a hell of a time. I appreciate a good challenge lol. i just got a fresh pack of blades so I won't have to cut such large chunks on the next couple transfers. Lol Everytime I tried to get that edge the myc would tear or slip under the blade. Also this is my 2nd time going back to spores on this print with a half dozen germ plates and I can't remember how many transfers. This is the most recent.
I do have enough gentamicin sitting around to pour another sleeve with it. Think I might do that. However it was pans that made me stop using it.they really don't care for that stuff. Think I'm gonna pour some more plates take a swab and streak more plates than I think it'll work on then hopefully I'll have a few relatively clean colonies to choose from.

Don’t bump your own question. If you would take the time to go back to the first page of this post, or the Pan Cyan Martha thread, you would find your answers. We’re not down for spoon feeding here, you’re going to have to figure out what works best for you on your own.

The recommended 25mg per 500ml. It's been a while since I've busted it out but the pan cultures I was messing with at the time were moving really slow despite temps being ideal. All my cube cultures were moving WAY faster and I couldn't seem to find that "wispy" myc no matter where I transferred from or how mamy times so eventually I made a sleeve w/o the gentamicin and transfered the same pan cultures to fresh plates. Some with and some w/o gentamicin. The plates w/o gentamicin really took off and hit the ground running and finally showed me some pretty looking pan myc. The plates with the gentamicin took a bit longer to take to the plate and continued it's slow pace while the culture that became nice, quick, and wispy on the regular MEA fluffed up, thickened up, and slowed back down when put back on the antibiotic agar. after that I stopped using it. My woodlovers also started looking a lot nicer on agar w/o gentamicin. Basically looked like the mycelium was more defined in it's structure making it easier to differentiate clean healthy myc. I also wasn't using peptone at the time. Maybe the gentamicin won't have as much of an impact in agar supplemented with soy peptone

Temps stay between 75-82ºF, just mist the lid and keep the one latch closed to keep the lid up for better air distribution, farthest gotten so far with pans indoors. Outdoors was easy when had the temps they enjoyed but thats no longer the case, this is just cased with verm. Pins keep popping out everyday so hope everything is being done correctly.

Ok so I have an lc question. I made a couple of those no pour lc jars with a vent hole covered in micropore tape. I've never really worked with it till now. Does it wick contams in like other filters if it gets wet? I have a practice lc started while I wait for these tropicalis spores to geminate. I have the whole thing wrapped in cling wrap besides a little where the vent can breath. I forgot to add a marble so I'm just being careful not to get the vent wet when I shake. I'll prob buy a stirplate for the trops but I'm trying to understand the limitations of this tape without scrapping anything...
