How do you guys select your mycelium when doing tranfers on Agar for Pan Cyans? With cubes I go based on speed of growth and thickness of the rhizomorphs obviously. So I have two different strains of Pan Cyans grown out on Agar. Jamaican and Australian. The Jamaicans look more thin and wispy than cube myc, but still fairly thick compared to the Australians. The Australians are so thin and wispy that it's hard to even see. I shot about 6cc from the spore syringe I had into 6 grain jars and after about 2 weeks, I thought I had no growth, but upon closer inspection, I saw that the entire jars were almost colonized, but it was so thin I could barely see it unless I got up close in the perfect lighting. Is my Australian myc unhealthy for being that thin, or is that normal for Pan Cyans? It looked exactly the same on Agar. I had 9 different cultures of it that I "isolated" down just with a single transfer. I've already used all my Agar wedges to knock up like 15 different grain jars, and they're all growing out already, so i'm not currently isolating, but what does normal look like for Pan Cyans?

If you are doing selections for me its just like selecting for cubes. Select for the fastest thickest, most uniform growth- to get a single phenotype that is healthy. The only way to know how well it will fruit is to take it to fruiting.
Australian phenotype tends toward thinness but if I were selecting i'd try to get it as thick as possible- Brain Bulb has a very nice looking Aussie colony a few pages back on this thread.
There is no "normal" growth form for Pan cyans- some varieties like Jams, Alabamas tend toward more tomentose and Aussies tend toward thin. There is NO relationship between this and fruiting ability. Some people may disagree with me on this one but I haven't found growth form to necessarily correlate with fruiting potential. I have had completely unselected mycelia (multispore)fruit well and beautiful monoculture colonies that didn't fruit well and ratty looking colonies that did fruit well. So far I have not been able to find any relationship between how nice a colony looks and how how well it fruits. These slightly discolored Jam colonies both fruited well:
 in this grow: https://www.shroomery.org/forums/showflat.php/Number/24820407
Both my Aussie and Alabama grows were from unselected mycelia.

3 days ago i dropped MS (pan cambo) on 4 plates. on one plate i did the same with a cubes. They are sitting around 68-70. I can see growth on the cube tray but none of the pans. do they need to be incubated at a little higher temp?

I didn’t have any luck with my Cambo MS. Wouldn’t take on agar and I ended up using the entire syringe in one jar. Took it forever to show any kind of growth on grains. I would go with Mary’s LC to WBS process.

Just to keep it clear the LC-> WBS substrate is Blue Helixes procedure.

My bad. Wasn’t trying discredit BlueHelix. After seeing both your grows I had to pick up some WBS today.:-). I got Pennington Classic WBS. What type do you guys normally use?

Some pan aussie. I have a tray that should be pinning in a day or two and two bags that will be put to trays on Friday.

These bags use "Blue Helix Procedure" but I used milo instead of WBS. Will probably switch to WBS next time because it will allow for more inoculation points.

@Asura
I’m just about in the same boat. Spawned on bag of Aussies this morning and have two more that will be ready in a few days! Mine were inoculated on Rye berries this time though. Just ordered some regular myco bags so I can try the WBS method.

I hope it's ok to jump in here and say that while going through my print collection, all from 2008 and earlier, I happened upon a Copelandia print. That's all it says. Well actually, it looks like "Cyan" was written first and then scribbled out. I'm sure it's at least 12 years old. I have agar headed to me, so I'm gonna try a few different old exotics.

Yeah, only got the one on the go right now, but I have several more jars of spawn that are almost ready.

Yeah! Most of that area is open for pinning except for the few spots where the overlay is especially thick.
