Hey everyone, happy to be here.  I've had experience with Cubes and have been working with agar as of recently with good results, but this is my first try at Jam. Pan.'s thanks to kind friend.  I know the mycelium looks different compared to the cube mycelium I'm used to seeing, how do these look?  I know theres a couple contams on the plates too but am curious if the white is pan mycelium and if it looks healthy enough to be transfered away

This is M.S. from a print, I deposited spores on both sides of each dish to make the most of my germination plates, thanks in advance guys :headbang3:

going to fruit them in a dialed in(hopefully) mini mono.. i also have a mini coolmist but im wondering if its worth the hassle.

Hi, Canharo,
Is this the first passage from the germination plate? These are nice and cottony.
If so its normal for this variety for the colonies to still look weird, sometimes you get a nice colony from the first passage (P1) but usually it takes more passages to get a nice symmetric one.

I wouldn't do just one of them, the phenotypes can evolve to something nice even from a colony that looks bad, ideally I like to have at least 3 nice plates to choose from. To increase your odds of success take all of them to fruiting.

ie. for plate 1:
From the top colony I'd take a wedge from between  1 o'clock and  3 o'clock. From the bottom colony i'd take a wedge from anywhere between 7 o'clock and 4 o'clock.

Plate 2:
Don't take anything, it looks like it might even be contaminated.

From plate 3;
I'd take a wedge from any where between  2 o'clock  and 4 o'clock.

Just take small wedges at the edge of each colony. Avoid the center.

if the plate on the left is clean id just take from that plate and toss the ones with satellites. no time for that.

Informative thread.
Im currently working on wild spore copelandia direct gill swab.
Unfortunately my cloning plates went to south being sloppy and since copelandia has thin stems.


They resemble oyster growth on agar.
This is 3rd transfers from spores swipe where yeast is present on the swiped plates.


Will post 4th xfer soon.
I can see 2 kinds of growth on my 4th xfer plates.
One thick and cottony radial. And others are flat thin radial growth. Planning to test this on few grains.

Hope you dont mind my 3 questions sister. I find your threads informative regarding this.
-I did open some of the xfered plates. Does copelandia have a sweet almondy smell to it?

-can copelandia plate survive the fridge for 3 months?
Im getting overwhelmd on agar work load asm
Working on 4 cultures eg. gano which cant survive cold storage.

-And when it comes to subs and casing is it really required for aged manure to be present?

Hoping to get my few spore prints to save so i can concentrate on this culture if time permits.

Got them id'd ,Very active. I got mind fucked eating few fresh grams as a reward after cloning and swabbing them. They turn dark/jet blue after 2 hrs working on em. See the stem of the 2nd picture thats 15 min after i plucked it up fresh but semi dried and sporulated.

Its cyan brother. Im not sure of the wild variety.
Its from grass field here in the tropics with 83f+ temp.

Hello,

I am looking for the absolute best recipe for pan cyan substrate..  I know this could be debateable.  Was hoping someone could look at the list of ingredients I have immediately available to me and advise what I should and shouldnt put into the mix.. Maybe a ratio?

  • Barley Straw
  • Horse Manure Pellets
  • Spent Coffee Grounds
  • Chicken Manure Pellets
  • Coir
  • Gypsum

Panaeolus bisporus Small Scale Direct Inoculation Grow

A PF tek/ Mac Merdin Pancake style grow. Except for the liquid inoculation its a super simple procedure.
I wanted to do a bigger scale grow using these:


but I couldn’t find them at the time, so I used 1/2 pint glass jars instead.

Acknowledgements:
MyColorado
MacMerdin

Path:
Colony on agar>liquid inoculate>PF tek jars>case & fruit.

Grow media/substrate recipe & preparation:
Compost* 500ml
Horse manure 250ml
Millet 500ml
Vermiculite 500ml
Azomite ** 1tsp
Garden fertilizer** 1/2tsp
Gypsum 1tbsp
H2O 500ml

*Compost was homemade
**The azomite and garden fertilizer were added as an afterthought. Azomite- mineral source. Garden fertilizer- NPK 5:10:5.
Added sub to eight 1/2pint jars (to the rim just below the threads)-> covered with dry verm-> PC 3hr@15psi.

Liquid Inoculate: Blender tek

Case- Peat moss: vermiculite 1:1, pH was brought to~ 7.5 with Calcium carbonate.

Procedure:
Print was germinated on ME agar

Pan. bis. mycelia colony on agar:


Liquid inoculation of the sub:
A suspension of the whole colony in water was inoculated- PF tek style into eight 1/2 pint jars. Each jar received 4x~1ml into the 4 injection ports.

Colonization of the sub:
Jars were incubated. at ~75-77F for 20 days while mycelia colonized the substrate.

Casing: Dry verm layer was removed. The case layer was added to ~1/4 inch- case layer was well dampened with dH2O.

Fruiting: In greenhouse-  at 76F to 79F and 80-88% humidity. [this level of humidity is about as  high as my wicking humidifier can go. I may have to supplement with an ultrasonic humidifier to get the humidity higher.

Pic two days after fruiting started:


A couple of the jars have fewer fruits but the rest are pretty consistent.

I'll add more pics as the grow progresses.
