I will bring a couple of photos from my last TTBVI tray. I didn't take a picture of it as a whole, but I have a couple of bunches that have grown on it.

I have a question concerning liquid culture.
When it comes to storing Cubensis liquid culture, it is advised to keep them at a temperature range of 35°F to 40°F.
However, I'm having difficulty finding a definitive answer for storing Copelandia liquid culture. Some sources suggest a range of 35°F to 46°F, while others advise against refrigeration altogether. I have searched through forums but haven't found a clear answer. Can anyone provide the accurate information? Thank you!

Germination plates from a TTBVI spore print (on grain water agar). Any advice on spots to pick transfers from? I assume the big, white blob in the 2nd plate is mold?

I got some pan cyan BVI on agar, wonder what you guys think. First transfer from swab plates. The black dishes are 1% ME, the crappy looking ones are GWA poured too cold.
Is it too early to say what to pick here? I guess the myc really likes the grain water. Is this growth too dense - should it be more wispy?

I had great growth on GWA, too. PDA not so much. Restreaked from spore print on Monday, heavy growth on GWA today.

When it comes to germination plates, I recommend a less-is-more approach. I do not cover the entire plate, that way there is room for the colonies to grow out before transferring. I only streak a single line, and a short one at that, right in the middle of the plate, staying away from the sides. I will streak the tool to 2 to 4 plates, that way it dilutes the spores and spreads out the colonies.
If you are starting with a spore syringe, I would really recommend using a sterile cotton swab to absorb the spore solution, then streak the swab, rather than trying to squirt a small amount onto a plate.
Your plates are looking very promising right now, but I would let them grow out more before taking any transfers.
Bismillah, all of your colonies are looking great, and those are some great photos as well! IMO, you could send the most consistent looking colonies to grain now, or let them grow out some more. Some of the colonies have some inconsistency to them, I would transfer those again. Nice work!

How is this looking for a T2 plate? Worth transferring from? Or back to spores?

Hard for me to say. I've never grown BVI, (yet), so I've never seen it on agar in person. It looks a little disorganized to my eye, but it might just be the way that culture grows, it could just be the agar, or it might just be the environmental conditions it's growing in.
I know good looking cultures on agar when I see them, but a lot of time I'm unsure. I would say try changing the agar recipe and take some transfers. You could try a different nutrient, or just change the amount of nutrient, and/or agar agar powder used. (I prefer agar as soft as possible and still be able to take transfers. From my experience, it has helped the mycelium's appearance, and contaminants have a hard time hiding on soft agar. If it's there, it will show itself undeniably.)
I will also sometimes make a small jar of grain to test something with. Once all of the new agar plates and grain test jars are prepped and cooled, I'll take some transfers, and send a piece to grain at the same time. It's a great way to learn when I have questionable cultures like that, and it diversifies my chances that something will work.

Thanks for the input! I swabbed three plates, plate B was water agar, the others MEA.
I store cube plates in the fridge if I plan to put them on grain later. Would this also work well for pans?

Interesting - and good - suggestion. I'm more of a (bacterial) microbiologist, so I'm accustomed to streaking for single colonies on a plate. It's just that, usually, my colonies don't spread like myc. Good news is that all the TCs keep beating the drum on spore prints, so that's what I have. I'm gonna follow your advice and do a short streak on several plates.

It might be hit or miss. Pans don't tolerate cold temperatures as well as cubes, (bisporus is an exception to that), and I've heard of growers having issues after cold storage. (At least from long term storage of LC in the fridge. Short term storage on agar might be different.) I wish had a better answer for you.
Since you have multiple plates, I would try some in the fridge and some at room temperature and see if it makes any difference.

None of this looks particularly good imo.
I typically use 3 plates for swapping and this has worked well for me.
With the same swab I will make a zig-zag across every plate. Typically, if I have any nasties, most, if not all of it, will show up on #1. Plates 2 and 3 will usually have some nice monocultures to transfer from...with plenty of space between.
