Happy hump day friends and neighbors!! I'm back to the agar. A few 1/2 pint jars. Gonna try a transfer in a couple days. This is after 1 transfer.... Gonna let it ride a couple days and transfer one more time (t2) and then if it's still looking good I'll put it on grain again and see what happens. Hurry up and wait!! I hope all you panheads are doing well. 😎✌️

Those don't look right for T1 plates. May I suggest you try actual petri dishes for agar? I used to try and use those jars for agar and I grew to despise them. The height of the jars makes it very difficult to not hover hands over the agar, and it is difficult to get the necessary angle to place transfers. Once I made the switch, I will never go back to agar jars.
I would also suggest stop using food coloring. I know plenty of growers use it successfully, but for now, I would leave it out.
You could use those jars for making BRF/verm pucks, and germinate your spores in those. You would still want to transfer a piece of the leading edge of growth to agar, (in a petri, IE, a real plate), and let that grow out.

I second that about the jars and even no pours as a whole. Grew to absolutely resent them. So much trouble with condensation, hovering, getting the wedge off the scalpel at an undesirable angle, cant see whats going on in them, mycelium wanting to grow upwards rather than outwards, etc. Can they be made to work? Yes. But to me it's just not worth the effort. Far better results much easier with plates.

I just ordered some plates. I've used plates before. I like to reuse my stuff. Finally found some I can reuse. Should arrive sometime this week. Thanks for the advice folks!! I hope you're having a happy hump day you panheads!! 😎✌️

For me, I like 1/4 pint Mason jars for agar. I use the sides of the jars to help with clean up. Plus, I can leave a jar in the box without any sort of wrapping or bagging and they stay fine. I just use MEA for most everything, including Pans. The only food coloring is just a hint of blue gel in my water agar to make it easier for me to keep track.

I did just that a few weeks ago, but maybe 1 drop was too much.. I can see through the agar, but have no clue if mycelium is spreading.. as it grows so thin, there is just the 'hue' of the mycelium against the light which gives a clue to how far it has spread. I'm struggling now to see with subtropicalis if I can move it to T1 already

Yes, for me, water agar took a minute to get used to. I understand what you mean by "hue" of mycelium. I recently started some Pan Cam 'Komodo' spores and it had such whispy mycelium.

I had that with with every specie I tried on WA thusfar. Regarding the komodo, I have it on rye and oats at the moment.. ran out of rye. But it colonises very slow.. no idea why .. I don't see any bacteria or other sign of contam. hope it doesn't stall on me. bisporus.. same batch of rye.. colonises just fine, maybe the oats are slightly on the wet side though (tried hamloafs oat tek, but got quite some burst grains..). What kind of grain will you use for the cambodginiensis?

Not necessary but good to add. Soy peptone, but it's not that fancy. Cheap versions work fine. There are also agar powder with peptone pre-added. If you're interested/like diy, you can make your own wheat/soybean peptone. (Acid hydrolysis)
EDIT: grain water contains some trace amounts of peptones.

So I'm also looking at potential LC for pans, I saw someone make their LC's (I forgot who - sorry) in a grain water style by grinding up their grain into a type of flour.
Im wondering if I could use millet flour from the indian shop in my LC seeing as I usually use millet as my grain? Or maybe even do a 50/50 with LME... I should really write stuff down as can't remember if I put 1g LME or 0.5g last time
Was thinking of trying 0.5gLME 0.5g Millet flour to 250ml water ... anyone tried this kind of thing?
Or maybe just go 1g millet flour hmmmm

When you all are looking for fast moving pans mycelium on agar, do you germinate spores on many plates or should you only germinate on a few plates, make a bunch of transfers from the germination plates and then see what grows out the quickest?
I've seen varying speeds between the two strains I've been growing(estero and BVI) but they haven't really been lightning fast on agar.

It is funny how the world works. I forgot I made a pint of oats for last night's cult work. A 1/4 pint of Peace River on WBS was split between pints of oats and WBS.
L-R low prep oats, rye and WBS (all Peace River). The oats were inoculated last night along with a different WBS (not shown). The rye & WBS in the pic were noced on 5/15.

Just a follow up pic
I had two of these. One went to grain as test.
But really, this doesn't look right. I dropped the plate (by accident) - on the desk. Bits came off the powdery stuff. I dont think I have seen any mushroom myc healthy or unhealthy do this. It just looks wrong.

What do you guys say?
(I have marked the jar that was noc'd with the other plate)
