If I manage to fruit the Ambrose culture again, I'd definitely like to share some spores around so that you guys can test them or send them for testing. It still blows my fuckin mind how something can be so potent.

Anyone tried to germ on brf pucks? What were your results? I saw it mentioned in the Josex Biopsy thread that these work well for pans.
Ive tried several times with pans while I was still struggling with cubes and never got anything to grow so now that Im a bit more experienced Im making another attempt to germ some pans on agar and also some brf pucks.

Sure I do it all the time to clean stuff up. Think about it - if pan mycelium will grow on GWA or MYA, why wouldn't it grow on a BRF cake?
What is your source? Spore syringe? How old is it?

Oh gotcha sorry I re-read your post and between your first and second post, I am now understanding that you just can't get the pans to grow from spore - it's not that you are getting tons of bacteria instead - it's just that the spores won't germinate right? If they won't germ on agar, I don't think there is any better chance of them germing on a BRF cake. We usually use cakes on bacterial spore syringes as a way to be able to transfer away from the bacteria.
If my understanding of your situation is correct - that you aren't getting bacteria on your plates - you just aren't getting germination, you have a few options: 1. Make softer agar. Use 16g of agar per 1000ml water instead of the usual 20. You can add peptone too - some feel it helps but I'm on the fence. 2. Mame a spore syringe and let the spores sit in it for 24 hours before squirting a drop of it onto a plate 3. Use the syringe from #2 above to squirt some of the spore solution (like 1ml of it) onto a BRF cake
If none of the above works, your spores may simply no longer be viable. That's not uncommon. Different spores expire faster and slower than others, and some of it also has to do with where/how you stored them. If you need a pan print, I'd be happy to send you a fresh one to get you back going again.

Are BVI actually more potent than other pans or is that just going around because of the one fruit that tested very high in the cup not too long ago?

Thank you Hindsight, much appreciated!
I have a couple different prints to try now, although I think the issue is most likely the thickness of my agar.

If you filter google searches to only articles before the date that was released you will see that no one ever implies BVIs are the most potent of the pan cyans before then. So I think it's because of the cup.
I think BVIs are the easiest pan cyans to get. Seems everyone has them right now.

Is there hope for this BVI clone attempt? I pulled these from stipes. Nine additional plates were bacterial, moldy or both.
