How old are the spores? How much solution did you use?
If you use too much solution it will have to dry out before the spores will take off

The print fairly fresh. I drew up approximately 6ml. Sterile water. Couple drops on each plate. I could see a couple spores in the syringe. Maybe I'm not playing the hurry up and wait game!?!? I'm new to agar and pans in general. I just appreciate your time! Happy Wednesday to you!! πβοΈ

3 days is not much, 1 week is more like the average and after 2 weeks I would start to think about trying again.
Next time try to inoculate your plates dry by transferring spores onto the plate with either a sterile cotton swab, an inoculation loop or just a scalpel will do as well.

Well, I have to admit, sometimes I mess up the ratio (e.g. amount of lme for 1000 instead of 500).. thinking I did it right. When nothing germinates.. happened 2x now, I just make some new fresh agar and give more attention to what I'm doing. Or my loop is still too hot.. just let the inoculation loop cool / dip in some fresh agar to cool it down well helps me. A well.. those are the things I tend to mess up.

I feel like this tray of Jambos should have pinned by now. It was cased 10 days ago and still nothing
In the past, I have messed around with a fan setup on a timer near my jcm. but I think every time, I've removed the fan and still got multiple flushes afterwards. Not sure if it is harder to initiate pinning than to keep a tray moving after it has already started.
I ran some ATL7s in this chamber recently with no fan setup and got good results

It looks like you could get some clean transfers from the gray plate, but the blue plate looks like mold to me.

If I remember correctly itβs fairly common to have issues with pan syringes for some reason. I think Jake and a couple others shoot it right to grain in these cases, and then put a colonzied grain to agar and go from there.

I posted this in another thread, but I was advised to re-post it here for better information.
I have some panaeolus cambodginiensis spores ready to go on agar or BRF. I have coir, dehydrated horse mmanure, and vermiculite on hand.
What is the recommended Panaeolus bulk substrate for these mushrooms? I've read that they like mmanure, but should I do 100% manure, 50% manure/50% coir, 50% manure/50% verm, 50% manure/25% coir/25% verm?
Will any of those combinations work? I've heard that they likee straw, but I really don't feel like going out and getting straw. I don't have the space to store it, really.
Also, I have Jiffy Mix to use as a casing layer. I used it in pasteurized form for some cube grows years ago, and it worked just fine. I'm assuming that it's okay to use for pans, too?

Jiffy mix is used by a few people and works just fine, it's basically peat an verm anyway IIRC.
I feel you when saying you don't want getting straw, I really don't like prepping it.....too much of a mess. For a while I used manure/coir 50/50 with good results and now I'm using sieved compost (contains manure ex factory) and coir 50/50 cause its easier to prep. One important thing is that you don't over hydrate the substrate so it gets soggy. Keep it a little bit on the dryer side for the colonization part and then you can give it all the water it needs once you put it into fruiting.

I am in central florida. I have prints from previous finds and I have cloned a stem sample to agar that I managed to get clean growth from. The prints I had lots of trouble getting any decent growth without total contam takeover of the plate.

Thank you very much! I'll keep the moisture content suggestion in mind. I feel like a lot of problems can be pinned down to excess moisture.
I've even heard suggestions to hydrate bulk substrate to a point where no water runs out when you squeeze hard, not just a few drops.

You can do that with cubes but not pans. Pans like more air.
I second Baba's substrate recommendation. 50/50 coir/manure is great. You can even go down to 25 manure and 75 coir or anything in between and it still is just fine.

Okay, I might have to try both. The 75/25 sounds appealing since I had trouble finding the manure to start with.
