ekabpils, great job! The two sections with different heights is neat, I haven't seen such clear distinction within the same tray before.

Speaking of funky looking bispo plates, check this one out:

This is from a clone. Probably a T1 or T2 I forget, but I took a few clones from my good bispo trays and this culture always turns yellowish and gets really thick in the middle. I haven't seen this before other than maybe pan cyans on really high-nute agar but this is not a high nute agar plate. It IS MEA though, which I reverted to for a batch of fresh plates out of laziness because I ran out of grain water and didn't want to cook more. So I don't know if it is the agar or the culture but probably the culture because another bispo clone on MEA is not doing the same thing. I'm sending it to grain just to see what happens. Also sending a more normal looking bispo clone culture to grain at the same time.

My Bisporus does that rhizo-yellowing-blueing on grains. Slightly on agar, maybe, sometimes.

Interesting! The WCRS I just grew did something similar on the casing once after a couple of days, I had never seen it before!

I just cooked up a batch of sub for some jars that are coming ready. Some are for my first bispo clones and am hoping to get a nice canopy out of them. Also hope to start my outdoor beds now that it seems warm enough. I'm trying a new sub material this time - instead of straw I'm using shredded hemp at VL's suggestion. It has no seeds. It's interesting stuff.... feels and looks more like finely shredded wood chips than straw so it will be interesting to see how the pans and bispos like it. I know that the mycelium like cellulose and some amount of nitrogen, and both straw and hemp stalks would have this, so it will come down to whether the pans like the denser chips. The mixture looks plenty fluffy to me, though not as fluffy as straw. I went with a 1:2:1 ratio of hemp:verm:manure. I think I read that Adas has grown pans on wood chips and HWFP before so I don't see why the hemp wouldn't work well.
I'll be sure to post the results.

I have grown Pans on sawdust-containing substrate. There isn't a single mushroom I've encountered that wouldn't colonize sawdust just fine, if it's leeched. It's really kinda like coir.

You can make a fancy punch like I did, and then not use it lol. All you need is a stainless steel thin walled tube. Easiest source for those is to buy a stainless steel drinking straw. They work fine but aren't ideal because they are thicker than they need to be which means they take longer to cool after each sterilization. If you want the best, get this: https://www.mcmaster.com/6100K356/ The wall is very thin and it cools in just a few seconds. Thinner wall makes for better cuts.
95% of the time the plug stays in the petri. You just lift it out with a blade. The other 5% it comes out with the tube and you just pull it out with a blade.

Why use the punch if you still have to sterilize a blade? I'm guessing that's probably why you don't use it lol I was thinking of making one but now idk. The uniform hole looks nice tho, I like me a good hole.

The punch I made uses the same stainless steel tube I provided a link for, but I added a handle, a plunger rod, a spring and a cap for the plunger. So basically on the rare occasions that the plug sticks in the tube, I can press the button with my thumb and the plunger rod goes down and pushes the plug out. All that added "stuff" (handle, spring, plunger, etc) isn't really needed and the plunger rod has a lot of mass and slows down the cooling after torching.
I still use a punch, but instead of the fancy plunger-punch, I simply use the stainless steel tube by itself.
Why use it if you have to sterilize a blade too? Because it's not about sterilization. It's about having a perfectly round plug to transfer, and the reason for that is that the shape of your transfer piece impacts the growth pattern of the mycelium. If you go with a perfectly round plug, you should get perfectly round growth. I want that because it helps me identify potential contamination, and also helps me see sectors better. It takes me 3 seconds to sterilize the stainless tube and 3 seconds to sterilize my scalpel using a butane torch. I then set them upright in a holder I made, directly in the flow of my hood, so they both cool down in seconds. I don't mind those extra few seconds. They are worth it for the benefits described above.
