I think the tray at the top left in this phost is the tray in your signature, could be wrong though. I remember being very surprised that they turned out so albinoesque.
Would be cool to see more recent photos and how these genetics develop over a few generations. Beauties for sure, hope the potency will come right.

Clearer pics would probably get a better response. It’s not necessarily a problem but I don’t think I’ve ever seen a ring around a transfer plug that looks completely transparent like your plates are showing. Could be caused by your nutrient formula. But I don’t see anything that would make me say these are toast, nor anything that would make me say these look fantastic. They are probably fine but I doubt anyone could say for sure. What agar formula are you using? For pans I strongly suggest plain old grain water agar. I get the most consistent growth and patterns with it.

Sorry Unicorn, looks like your first post got missed earlier. Welcome! Yes, you are in the right spot.

The Nec D looks great and I would take it to grain IMO. The Peace River might need some more cleaning up. My print of PR was kind of dirty, it took me a little luck and a lot of attempts before I got a clean culture. It had the same kind of look before it was cleaned up.

Thanks a bunch and yes there were some white spots that grew and turned brown or black in the t1 and t2 plates. They are looking much cleaner now. I am using dry malt extract. I will give grain water a go.

ah yeah, time to see what these wild coast red spores will throw out. Fingers crossed for something worth cloning. And toes crossed temps stablise a bit, I've got things as insulated as possible but the night times have been chilly.

Remind me Coversall, are all of those from a single culture or a bunch of different transfers from MS? Basically asking if you expect to see a variety of different things happen with those jars.

I've got 15 jars that I nocd with 5 different LCs started from different plates. I'm not super knowledgable on this side of things, but my aim was just to get as many different culutures growing with the limited space I have. In the hope that will increase that chance of something good growing... I could be way off the mark here though. At the end of the day I really like the tiny jars haha

Got it - that is what I thought. You are doing what I usually do which is to germinate and then take a transfer from multiple different nice looking germination growths. Keep them separate and transfer each one as many times as needed to ensure they are clean, then go to grain. Each plate-jar-tray will then likely produce something a bit different so you’ll have a good chance of finding something good. Vs just picking one plate to make LC from and knocking up multiple jars with it. The other way I like doing it is to put like 4 unique growths from a germ plate into a single plate and let them all grow out. If it looks clean I will put the plate to grain. That way I’m getting as many genetics in as possible. I’ll often make like 5 T1 plates like that, from a single germ plate, with each of those 5 plates having 4 distinct cultures in it. The next thing I will try is making spore syringes off my own sterile prints and putting them directly to BRF cakes - no agar or grain - fruit on the cake. If it works out well it should be the fastest way to stabilize something.

Yeah that's pretty much what I am doing here. The LCs were mainly made to have clean, injectable solution. It could easily be an LI rather than an LC. But LCs have been working for me so I don't want to rock the boat too hard.

I wonder how well pans would fruit off of plain BRF?

Is the main component of this idea starting from spores?

From what i have read, Pans do poorly on just BRF but if you can get even a single fruit to print, it might be useful when you need to conserve agar.

Unless u meant Pan-Cakes :cool2:

Yes - I had planned to experiment with various formulas of cakes and maybe come up with some new ones of my own depending on how well the established stuff works. I have gotten in the habit of calling all cakes "BRF" hah.

Hindsight, how are you getting sterile prints with pans? Cubes are
easy to grow in vitro but haven't seen a sterile pan grow in pods yet.

When I say "sterile", I mean sterile enough to have a print unlikely to contain contaminates. Basically what I mean is to use the method where you cut your foil squares for your prints, line the bottom of a shallow baking tray with them, lay another sheet of foil on top, set a wire rack on top of that, then cover the whole thing with another sheet of foil and bake @ 400F for 30 mins in the oven. Remove it, put it front of your flow hood, peel back the top foil covering and set your caps on the rack. Re-cover the top with the foil. After 8 hours, you put it in front of the hood again, remove the foil cover, remove the single layer of foil that the caps just printed on, then recover with foil again, allowing the caps to now print onto your foil squares. Any contams on the caps would have fallen off onto the first sheet, so now it should just be spores. Everything else in the process from the pan to the foil to the rack is sterile. Obviously there is still some risk since the caps themselves are not sterile but the first print should take care of that.

Ok, cool.

I was thinking of making some PF Cakes, inoculating with spores, colonizing the cakes then crumbling them with a manure substrate into a shoebox set up. Or maybe i'll just go with the manure-based Pan-cakes. I still havent descided yet....
