That’s cool that they survived! I’ve got wbs jars of bisporus and cyan I just started from ms. They’ve germinated so that’s a decent start 😅

It doesn’t seem like the pans like wbs and or how I prepped them. Maybe a little too dry, I live in a humid climate so I figured drier prep would be wise


Here’s bisporus (WA variety) myc is on the bottom front edge of the jar



Here’s cyan (Hawaii) I’d be surprised if anyone could find the myc. It’s growing off just a couple of millet toward the top



It’s just been like 5 days since inoculation. I’ll update when I have more substantial things to show. Just wanted to get the participation ball rolling

I live in Hawaii close to the ocean and my windows face the east so I should be able to get decent results with unmodified totes/containers placed near a window

What up peeps.  I’m working on some small pan cyan tubs for post ssri taper :smile: and have some questions for you pros out there.  Picture 3 has not been cased and is at 9 days since spawning to bulk.  Is she ready to case or keep her going?  Pictures 1 and 2 have been cased, about a week ago.  How would you address these blobs and lack of pins?  New casing layer, mist more/less, less/more FAE?  How many times a day are you misting before and during pin set?  Are these signs of being over field capacity?  Thanks and mush love!

Are you misting your tub during colonization? Looks matted.

Never had blobs like that and that is a bit more yellow than I'd like to see, probably reaction to bacteria.
Hard to say what went wrong without some info about your process.

How have you prepped your grain (PCed yourself/premade bag)
How have you inoculated your spawn (agar/spore syringe)
What's your bulk substrate and how have you prepped it (pasteurized yourself/premade)
What are your temperatures.

Have you got photos of cultures and such?

Spawned from MSS to multi transfer agar to wbs to steer pop coir per your tek.  Used the stuff found at Home Depot.  PCs grain 75 min, sous vide sub 4+ hours.  I think I may have been a little wet with the bulk.  Honestly have gone pretty light on misting. 

How’s the third one look?

Your sterilization/pasteurization times are both a bit low. Min. of 90 minutes @ 15 psi and for sous vide pasteurization I do minimum of
8hrs. I prep the manure on the wetter side when pasteurizing cause it helps to transfer the heat better but then have the coir dryer so it
ends up at field capacity when mixed.

Never used steer manure, I imagine it being quite muddy. Might needed some more coir or some verm. Soggy substrate can mess things up,
happened to me when I first used compost.

The surface on the third tub looks unusual and not quite there yet but 9 days should be long enough so IDK. I see quite some
yellow/metabolites there so might just go for it, I doubt it will improve much with more time.

There is definitely something not right with those tubs and I have no idea what went wrong or if it can be fixed but since you have three to
play with I would do exactly that. Increase FAE on the first one, re-case the second one and hold off on the misting for a few days and
case and max FAE on the third one with a bit of misting......stuff like that :shrug:

Time to start some new plates and take some photos along the way.

Just further to Adas’ observations. I germinated my Pan Jambo swab just fine and it had been in the fridge for 11 months! Also when I got germination I put that plate in the fridge for a week or so before I read on this thread that it was possibly detrimental. Have since transferred from said germ plate and got good growth.

Nice, Hawaii is the ideal habitat for Pans! You have a Washington Bisporus variety? I wasn't aware of them being native there!

I've already proven that storing spores in a freezer does no harm (see my journal post about freezing spores), so it doesn't surprise me that your swab did fine. What surprises me the most is not necessarily that the mycelia survived some freezing, but that they survived MANY cycles of freezing/thawing.

P. cambodginiensis Jambo (2020 print) - Everything looks powdery/not good, so I will not take transfers unless you all direct me otherwise. I think I need a fresh print of this one.



P. cyanescens Weza (2022 print) - Some good spots on plates 207-4 and 207-6, yes?



Panaeolus × Huastralian Venom (2020 print) - The large spot on 207-9 looks most promising. The other plates have a couple little spots that could be ok?

Moopers, for a beginner it would greatly help if you just used some standard Agar formula. All these colorants can make the myc struggle and look weird. I suggest you to try normal media (like grain water agar, no additives) and see what that looks like. The colonies look weird, I have no doubt there is some good genetics in there but the media itself might prevent them from showing their character.

Thanks. Searching around, I see that avoiding colored agar was advised a few times in this thread in the past. Neither of the pan teks I consulted advised against colored agar, though, so it wasn't on my radar. The color hasn't been an issue for my cubes, nats or tamps, so I wouldn't have guessed it would be a problem. Damn!

Another factor for the odd colonies may be that the plates with the most powdery/least promising colonies are from 18- to 19-month-old prints. The Weza print is recent and has some decent-looking stuff on 207-6?

The only other plates I have are dyed blue or are carbon agar, and they're all pre-poured, and I have 20+ plates left. I've reached out to my vendor to ask for the dye ingredients list.

I will transfer to the carbon and see if it's any better. My previous grow attempts of Pan cyan Goliath and Jambo, the carbon agar did fine. Though I don't like to germinate on carbon because I can't see the spore clumps to streakthem/spread them well.

Ah yea, you won't be able to get high quality mycelium from such old prints. The 207-6 is probably your best bet.

There's a few things I might nab from 207-3 just to see how it progress on a clean plate. There's plenty to take from on the Weza plates. 207-8 on the upper has some stuff I'd be tempted to transfer again to see how it develops, and 207-9 has that obvious bit that I'd probably transfer to.
Like Adas said I'd be transfering to a plain and simple, uncoloured agar mix. Just to rule out anything fucky.

Explain, I can see how getting an older print to germinate can be a little difficult but why after germination would the myc not be high quality?

Probably DNA damage. Who knows, but I've always seen that old spores produced weak mycelium.

Where do you get powdered straw, or do  you have to make it yourself? I suppose you could do so in a coffee grinder or use straw pellets - I've been trying to find pellets for a while now but they're continually out of stock.

I'd imagine if you DIY your straw that it'd be quite labor intensive.
