Wild print pan cyanπ def super thick - the other two trays the spawn went in did not fair so well. Sparse but also thick.

Why did you germ in LC, did you have trouble on agar? I'm having issues with a pan print myself, currently hot poured a thin layer on top of 10 ungermed, 8-day agar plates in an attempt to trigger some action. Will report back if anything happens haha.

I've got a spore syringe so I just squirted some into 12.5 ml of 0.4% ME LC broth. I was trying for a few drops but fumbled it and got a lot more (1/4 ml?). Flamed the needle red hot before blasting of course.
Yeah there's something funky with my agar at the moment, still figuring it out... Even cubes and bacteria aren't happy on it. I was getting plates either drying out or going bacterial, and even then bacteria didn't do so hot. Current suspect is city water or the premixed agar I was using. Got a new batch of from-scratch agar sterilized but waiting for a lower humidity time to pour it.
I got the idea of trying LC germ from another thread where people were brainstorming how to get around agar germ woes. That pic I shared is after sitting about 12 hours; lots of spores germinated. On the other hand I didn't get anything on 5 agar plate attempts. We'll see if this LC comes out clean enough to use!

i had issues with mine as well, you could literally see a sharpie streak of spores on the agar and it took forever to germ. After a week and change with nothing I took a transfer of the densest part of the streak and smeared it on a new plate and that germed before anything else. I think it helped the first plate germ as well from the melted agar from the hot scalpel because it germed from the transfer area not long after taking it. I hail mary'd and jammed the swab into a plate and that eventually got some germination as well. Quite a few T1s later I finally have a few T2's that look half decent, hoping some T3's will be ready to go

Monoid, I would suggest putting a drop of your solution to a few plates to ensure cleanliness before going to grain. Hopefully your new agar will work.

Interesting, I have managed to heat-germinate some spores before. We might be on to something here. But I tend to just heat the plate from underneath with a lighter. It didn't work with Subtropicalis tho.

Ooh thats interesting, now that I think about it, smearing the spore trail with something hot might have been simpler and more effective than the hot pour I did...hopefully I haven't made it too thick and oxygen can still diffuse through...So how long did germination take in the end, do you remember? BTW I had no germination issues with the bisporus, took 3 days. Only the cyan is making me nervous That 4 to 5 days is the critical period, if I don't have germination by then I just know something is probably not good haha. But a few members did mention a 11-12 day germination period, so who knows, I'm still hopeful.

Oh, that's significantly warmer than I've been keeping em at. I'm at more like 21 degrees C.
