Thank you all for your comments about the NecD potency. Fully aware of the SSRI thing, and not everyone has a hard time with “fasting” for a few days (I know some Coke’s who get painful “zaps” and others who just get grumpy or sad.)… but it is not a very good solution anyway because there are actual structural changes in the neurons (receptor sites) that take much longer to reverse.
Jake thanks for the heads up re storage. I have been treating them like cubes - dry well seal in a zip lock with a desiccant and call it good. Are the pans more susceptible to oxidizing because they are so small (higher exposed surface area to interior area ratio).

One thing that isn’t terribly clear from the abstract (I have not read the full text) is if the fresh fruit bodies stored at - 80 C were flash frozen while still hydrated. If that were so, it makes sense that would be the worst preservation of alkaloids - all cell walls would be utterly destroyed and upon thawing the goo would expose nearly all the alkaloids to oxidization. In the second part of the sentence they do describe the fruit bodies as “dried biomass” so maybe it is safe to assume they froze fresh un dried fruit bodies…

I don't think the oxygen can react with Psilo without any moisture present. I've had some 2.5 year old Serbica, where 1g kicked both of my friends' asses (1g each).

This is a wonderful idea actually. I've thought about modifying the lids with a valve and using a vacuum pump to remove the air. But your method is way simpler and faster. Hats off.

PsiloPsychin, I read what I could, but I don't have Microsoft Word so some of it may have been missing. It definitely wasn't clear how they were comparing dried biomass to fresh biomass.
I started questioning everything they did when I read they used a "sterilized terrarium", and had problems fruiting cubes! They apparently didn't understand air exchange and evaporation and how that affects fruiting. (For the longest time, neither did I, but I still got fruits.)
It's still an interesting link, thanks for sharing!
Adas, I'm glad you like the vacuum sealed jar idea! If you try it let us know how it works!

At first I was going to buy one of these foodsaver accessories for jars but I looked at doomsday prepper vids on YT for inspiration and now I vacuum seal my jars in a small vacuum chamber I've build, lids unmodified.

I found a pdf of the entire report. The highlights are not mine. Regrettably, the precise method of preparation of conditions remains unclear even with this additional detail. I always tell my students that if another scientist cannot precisely replicate your study by reading your method description you have failed at writing up your method. I’m giving these guys a C for their method description. Inferring from information scattered about in this report, it seems to me that the samples that they stored in the freezer were first diced up (see section 3.5). If I’m correct, then these folks also introduced a confound into their experiment because conditions 1. & 2. were dried whole (or nearly so) then powdered and conditions 4. & 5. Were first chopped up fresh then dried and stored. So the comparison of 1 & 2 isn’t just a comparison of storage temperature it is also a comparison of processing / sample preparation. Oddly, they comment on how much chemical change is occurring during the chopping of fresh mushrooms but fail to incorporate that into interpreting their results. In addition, despite commenting on considerable variability in alkaloid concentration from one fruit body to the next … they make no effort to account for this in their analysis … for example, by including more than one fruit body per condition, or by random assignment of fruit bodies to condition and replicating the pattern of results in repeated experiments with different fruit bodies. For experimental design I give these folks a D. There is still some information here that is useful… I am intrigued by figure one and the implications it may have for drying temperatures. The old cap versus stipes concentration / potency difference seems supported but they do point out that the comparison is not significant due to variability in individual fruit potency… once again these scientist types might have benefited from talking to some cultivators 🤪

Yeah idk I do know sometimes I’m lazy I don’t vacuum seal them I just put them in a ziplock and I can absolutely notice a potency decrease compared to the vacuum sealed fruits, of course fresh is always the strongest

I read that article a few month ago and was disappointed. Seems like they didn't really planned this through before they started. I had to do a few test series on material properties in my studies and it took me a few attempts before actually getting usable results because there were too many variables in the process that I hadn't had identified beforehand and in the end I ended up comparing apples with oranges.

The first couple of clone tubs are moving fast. Colonized in 5 days at my usual spawn ratio of 1:2.5. I think they are ready to be cased as they are pushing out metabolites already.

Got more tubs following so I might just give it a go.

Hello everyone.After a while, I'm back in the ranks of the pan-gentlemen.Today I was pleased with my fruits Nec D.Thanks to Jake for this wonderful hybrid.There were questions about the potency of this mushroom above.So-he is incomparable with any of the pan and is very strong. 2 grams are carried away beyond this universe.

Hi Evertking.
I'm not JCM, but I wanted to welcome you to the thread.
I don't want to speak for JCM, but just in case it takes some time before he replies, I will. You may have read that he lost his brother recently, so he hasn't been able to spend much time here. We are all looking forward to his return, which according to him, shouldn't be too long.
I have read this entire thread and just about everything else I could find regarding growing pans. AFAIK, nothing has changed with JCM's fruiting chamber. He has talked about setting up a Jartha tent if he gets a bigger place, but only to have more grow space for additional varieties, tests, etc. If you only want to grow one tray at a time, the JCM FC is an ideal method.
Don't overlook Baba's pans in monotubs, the link is in his signature.
JCM, in case you read this, please correct anything that needs correcting. I'm only trying to help by giving you time until you are back to speak for yourself.
