I think you may have just saved me a massive amount of headaches with that.

I have to make up some impromptu agar plates tonight, my new dishes won't be in for about another week. I should have at least a few ready by morning to start the process.

This plates were bacterial as fuck, I wanted to throw them away.
Thanks to tip trippy I checked them today and there's some nice growth. Just gonna wait a few days to transfer it.

With him mentioning that, I started looking through my bookmarked pages. I found this a while back and haven't really had the opportunity to try it out. I'm not sure yet what would be used as the sterilized coverslip side from a small piece of foil.

Quote:

Contamination control and removal are very important technical aspects of microbiological research. Bacterial contamination is very common in fungal cultures. Currently, the commonly used approach for inhibiting bacteria is antibiotic treatment; however, there are drawbacks to using antibiotics, including incomplete removal, limited antibacterial spectra, tendency toward recontamination, effects to fungal strains, and potential risks to the environment. Therefore, in the present work, we developed a new method for bacterial removal from fungi cultured on solid medium, the Cabin-Sequestering (CS) method, based on the different culture characteristics between fungi and bacteria. First, 3–5 mm round or square holes (the “cabin”) are excavated on a solid medium plate. The fungal strain containing possible bacterial contamination is inoculated into the cabin. The cabin is then covered with a sterilized coverslip, followed by incubation at the appropriate temperature. After 7–10 days of culturing, fungal hyphae grow out along the edge of the coverslip; however, the contaminating bacteria cannot pass through the space formed between the medium and the coverslip and, thus, remain in the cabin. The newly grown fungal hyphae around the coverslip are re-inoculated into fresh culture plates, where they form bacteria-free fungal colonies. The CS method is easy handling, with a short experimental cycle and rare recontamination. When necessary, it can also be used in combination with antibiotics in bacterial removal operations.

Yeah, there's a thread in the advanced subforum. People here pour Agar on top of the sample as a coverslip. Seems to be the same principle. Stall out the bacteria, let the myc run wild.
But if it works without this, especially for a fast growing myc like pans, I see no reason to use this.

That’s a beautiful tray man. If it’s Cambo that seems to be fairly normal for spore grows with decent conditions.

Those caps should grow significantly over the next day or two I think.

I’ve also never gotten a clean pan clone. Just gotta wait for the myc to take hold and run past the contam.

I am following Asuras write up:
https://www.shroomery.org/forums/showflat.php/Number/25987049
and following his tek.
He sterilizes his substrate and then knocks it up with LC.
My bags have been colonizing for about three days, but I am getting a brown liquid.  Is there any reason for concern here?

I am not familiar with using sterilized manure sub. However, I am using a new batch of horse manure this go around (instead of my bagged store bought steer manure) and I am noticing dark colored metabolites. If I had to guess, that's what your brown spots are. Continue as normal and see how it turns out.

@Angam such beautiful work as always! Love your posts

I was always under the assumption that you didn't want sterilized manure only pasteurized is that no longer the case?

If I understand correctly, the difference is that Asura inoculates sterilized manure-based sub in bags with LC, waits for it to colonize fully, then births this to the tray, and from there the process is as usual. So, the sterilized manure sub doesn't touch air until it's fully colonized. Asura doesn't spawn colonized grain to sterilized manure sub, which would increase chance of contam. Asura or others who know his process can correct if my understanding is incomplete or wrong.

Just ate some stones I grew for the first time and have started to realize what I’ve been missing only growing and eating cubensis.

Anybody here willing to shoot me a print? I have a fair variety of cubensis I can trade, but haven’t fruited my Tamps (yet).

Thanks man! Much appreciate your feedback.  I considered also that it could be extra water displaced from the bag that the mycelium did not need, much like grains that are over-hydrated, but prefer to err on the side of caution.
