these are pan cyanescens the first transfer from spores. On grain water agar. Maybe two weeks old. If that

Alright, well this is expected for just the 1st transfer. Like I said, sometimes my cultures are also wispy that early in the game.
Also change your media. The media could also have an effect on the visual appearance of the mycelium. Grain water is a hit or miss as far as accurate measuring goes (Strength wise) your media could be too rich,
Try out something more readily reliable like MEA or even PDA. If you don't have a good sugar source you can use potatoes by them selves. Most cultures don't mind starch alone without an added sugar/dextrose/sucrose.

All I really have is pda. I try to only use it for germination because it isn't the easiest to spot bacteria on. But you're right it'll be the easiest to measure.

What did you use to transfer into your agar? MS, tissue, spores/print, and also how did you transfer them in i.e. - drop out of a syringe,inoculation loop, swab? Just curious as I'm pc'ing my first set of pastyplates and starting up some agar work. I've only done it once before and it was shooting a ms syringe onto brf agar substitute, but want to get nice clean manageable growth to isolate. Thanks

Thanks, I've checkout out those threads as advised by another member. I was just curious about this paticular plate and method used. When i start mine i only have ms at the moment so i settled on using an inoculation loop i made out of nicrome, flames sterilizing, then into the sab to squirt a drop or two to quench the loop and then another to load it before dragging it on my pasty. Any tips? The only other plate i made by shooting a drop of ms through a ship onto a plate made of brf agar substitute, but i like the idea of smearing with the inoculation loop on the plate for more isolated growth. Reguardless thank you again for pointing the way as reccomended by bodhisatta On spores and agar
Doing this dramatically helps your chances as well as softer agar

Gonna bio those bisporus tomorrow night! Im so excited..im going for 1.5 out the gate and then prob up from there...i may redose with cubes though to experience the combo aswell

Check out my Cinctulus plates guys! They have recovered after the transfer and are doing really well. They are so wispy and frail looking but bright white and B-E-A-UTIFUL! haha
