Once you’ve had some practice and you know what you were looking for you can rather efficiently blind sector quality genetics it doesn’t always work out how you want but I am pretty good at spotting good pan strains on a dish. However there will still be multiple cultures in these transfers and I will still select a clone from the grow to create my isolate ultimately though this can just be done from a MS grow and you will get the same result it just might take a little longer to find that perfect clone candidate

Thanks Jake! Hopefully I'll get my eye in. I've got a little collection of pan prints to work with this year so we'll see how I progress.

I'm a strong believer in doing selective multispore cultures for my first run or two till I find several good clone candidates because just cuz a clone looks good doesn't mean it'll be a good culture. So I take several and run several small grow to find the right one which is basically like what jakes talking about . Otherwise blind isolation just causes more heartache than good.

I think that depends on transfer size and how early you took it. For example if you transferred a piece equivalent to 10% of the germ colonies perimeter to a T1 plate and then 10% of that to lc then you would have approximately 1% of the original germ colony, in that LC and whatever genetics that provides.
That is very rough mind and doesn't account for xfer size vs colony size, varying sector sizes or different growth rates in the genetics.
You do lose a lot of genetics with each transfer. I'd confidently say way more than 75% each time, but you'll still have tons of genes to play with and with pans a lot of the genetics seem to be trash anyway
(...well I've germed more unusable than usable myc with pans, probably not true of everyone, I dunno.)

I'm at a loose end waiting for stuff to colonise so am going to case and fruit four jars I had half forgotten about. They've been sat fully colonised for, jezz, almost a month.
Three pan cambos and one pan cyan. On brf, manure, chopped straw, and verm.
Any bets on whether it'll fruit or not?
I was thinking about soaking the cakes over night seeing as they've sat for so long.

yall are overthinkin it way too much if ya ask me lol
just streak a plate then transfer some germination points to new plates and let them grow out a tad. then transfer the desired growth from those to new plates then grow those out and use them for inoculation if they are clean. i dont know where or why it has become so common in this hobby for ppl to wanna spend 6 months on agar before fruiting a ms culture. im with jake 100% get urself some good lookin clean growth as fast as possible fruit that shit out and select some good fruits for clones then from there you can work it down to where u want it.

I was just wondering how much I've reduced the gene pool by going T2 to LC.

Your probably not wrong. Although I have yet to go past T1, I've just had some really shitty germ plates and tried/hoped things would work that didn't. But hey ho, bit by bit.
I have 2 good cultures now. Gonna take one to grain later, so we'll see.

ya i usually will take 4-8 if i have what i like. i took 6 from the last tray of necs and got 4 on grain now the other 2 looked weak so i didnt use them. when i find a good clone tht has potential thats when il start taking lots of transfers off it and fruit them all out to try n get it the best i can for slantin b4 keeping

This. My last few grows were inoculated with wedges from the inoculation plate and I had good results. I don't even bother cloning anymore. And for my next grows (from my own prints) I think I'll just scrape some spores in those cakes I make.

Soaking won’t hurt but pan culture likes to move fast I know once my grain is fully colonized I only have about a week to use it before my result start to drop off significantly if I get anything at all

bisp myc looks a tad different tho just so u kno. heres a cpl shots.

u should have ordered prints tho man. youl see why when u get them on agar. prints are a lot easier to work with and are way cleaner
