I've had that kind of growth too. I don't think it will work out.

Finally got the jambos down on some soft agar. I could barely touch the agar with my loop without hacking it all up haha

I did that a couple times. Make sure you are 20% agar-h20. For me the the main culprit was not swirling the bottle as I went. Causing the bottom of stack to be liquid and the top hard. Now I swirl the bottle frequently and as I have fucked up many plates. It’s best to NOT pour if conditions are NOT correct. When it hits the plate, it’s too late.
The other day I was melting agar from the fridge to pour 2 plates. I did not have enough to do a stack, not enough time to make more and I really just needed a couple plates for transfer. I let it cool too long and I got lumps. :takingnotes: Make EXTRA!

I’ve never used a loop, just a swab. I imagine the loop would be best for soft/wet agar. I’m sure the jambos won’t mind.

Yeah man I made this batch of agar softer for germinating spores. And I poured them thin. They are 8g agar and 10g lme

I have some sterile swabs on the way from Amazon and I couldn't find any locally or I would have used them

Yeah me too... the loop never really worked for me. I love my sterile swabs.

Same. Swabs are cheap. I never could get the feel for the loop.

FYI, never had a problem germinating on harder plates. But all the prints I use are pretty fresh

I have a crapload of prints that are ~7-10 years old. I’ve only gotten one Pan. cambo to germinate and the rest have been failures. I’m really hoping softer agar will help but I’m not super hopeful.

Man you should PM Mycolorado. I forget the details of his method, but it's along
the lines of cutting up a section of print and putting it in a vacutainer with
sterile water to germinate. Forget if/what are the nutes in the solution.

I think I’ll have to... I’ve got a couple I’d really like to revive. They’re both old Alan Rockefeller collections, one is a  P. mexicana and the other is a P. zapotecorum. The prints are pretty faint so I’ve been waiting for the right time to try to give them a go as I don’t think I have enough to try many more times.

The wc print I got seems to be getting dry, crusty. I was wondering if they would germ? I made sure to streak as soon as the hot water (Pyrex) bowl was cool.

Just checked them. Wc has germination ahead of the jambos I did same night 1/19. Actually none from jam yet. Wc is on its way!

I will take pics of my jam t-1’s later. Some good things I seen at a glance.
