Hello!
I'm just wanting to know if I have finaly had some success.
I know that this was a bit early to go opening petri dishes but I wanted a better look at what was inside.

So waht do you think? Is it myc or mold?

In case it's relevant, this is 7 days after inoculation on MEA

I get similar looking spots on some agar plates of mine, I can't exactly tell you that they're not contamed yet because I haven't seen it developing. I'm going to assume it's pieces of mycelium that touched the agar below or when you were placing the inoculant

Separate this dish from the rest. The contams that landed on the dish after you opened it will grow out. There is still a slim chance that mycelium will grow, but you'll have to carefully take a small healthy sample and transfer to another sterile dish. Keep an eye on the other ones. It's too early to tell if it is mold or not. :goodluck:

Thanks Psilosoulful, you have helped me a lot so far. I was going to place the dish back with the others but now I have it re-wraped in parafilm in a separate drawer.
I'm not fussed about this dish getting contaminated because I have backups but I'd have been gutted if it contaminated my other dishes.

I'll post pics up again once I see some more development.

when u take plate pics, hold it up to the light.  u want the light from ceiling fan/etc to be shining on TOP of the plate.
if u light it from behind, the texture of the mycelium can be hard to make out


when u don't hold it up to the light, the flash will make a glare, or if at the wrong angle (like in ur picture) the glare will prevent good visibility


this is ideal


worst comes to worst, put it on a white piece of plastic (like a tub lid or white cardboard) and take the shot from straight above, with no flash

if thats from spores and you got same amount of growth on other plates
its time to do some transfers (yesterday).

OK thanks Blindingleaf, I'm afraid that the auto focus on my phone won't let me get any better shots than this for now. I'll try and get better with a decent camera ASAP

spacechildo, this is the very first sign of growth. There doesn't appear to be any rizomorphic growth at the moment, just fuzz  with a cottony lump at the center.
Where would you transfer?

Any Contam on this Jar? when i look close at the myc in person the dark spots look like the myc hasn't gotten thick enuff to cover the colour of the substrate.. but i could be wrong..

It'll cover the substrate, just give it some time. And take that foil off, it traps moisture on the jar lid, which creates a perfect climate for contams to grow.

transfer from the leading edge. dont bother looking for rhizomorphic mycelium, its just a reaction to lowered nutrition in the agar recipe.
what you really want is to just get clean myc. by transferring clean myc to a new plate you hopefully leave any potential contams left on the old plate.



why is there foil on the jar? and how big is that jar? looks good so far IMO.

you have a dry verm barrier dont you? that's your filter from contams.
dont disturb this dry verm barrier, it needs to sit tight to do its job so you shouldnt really pick up pf-jars.

If you have a dry verm layer(which you should) contams will get trapped by that layer and will be unable to get to the brf/verm mix.
