Colonization problems

Hi guys, I have a failed colonization of GT in Rye jars. I made up 7 jars, used about 10cc from a spore syringe, left for a month and slight mycelium growth only in 1 jar, which ceased and now seems dry inside. The other 6 just did nothing.

So I prepared more rye grain, used a second GT syringe, this time had the grains many different levels of moisture, and again only 1 of them had some growth.

I took the first jar which I had some growth which died off and I did a grain to grain with some moist rye. Still early days in that.

I have them high up to try keep them warm, they are in a box to be dark. I have used 2 spore syringe and feeling a little disappointed now. I only need one to colonize.

Any advice?

This is the kind of problems you see with going from MS straight to grain. Rye is easier to work with and is easy to get spot on moisture content when talking about grains. I dont think there's much you can do at this point. MS man, its just a crap shoot :shrug: especially when going straight to grain.

Room temperature is fine...you don't really have to keep them warm... And I don't keep my jars in the dark...
Expose them to indirect light from day one and they will actually colonize faster...keeping them in the dark is outdated info.:goodluck:

Next time use a drop or two not 1cc spore syringe. Way too much.

Don't give up. And maybe order some agar so you can see the growth first before putting it on grain. Could be done simultaneously while you try with syringe to grain (starting petris) this will let you isolate clean and vigorous growth drastically increasing myc's chance for success

Either you've got bad syringes or your rye is horribly misprepared.

1cc per jar will do fine.  +-50% won't matter.

I haven't used rye personally, but you should detail your preparation, jar setup, and PCing.

Still, even with shit preparation, I'd expect more germination than that.  Probably the syringe.

I spawned some popcorn casings and had double-overlay cause I didn't put enough hydrogen peroxide in my automated aquarium mister.  I only got one mushroom so I cut off the head part where the seeds fall from and put it in a jar of LC and sprayed it all over a tin of PF cakes I made with gravel, cardboard, and bisquick in my microwave.  I think it will be good cause B+ is so potent.
Triggered yet?

Hopefully not too quickly :evil: The reason we don't go from MS to grain is because we cant tell if there are hidden contams. Not that it wont work. It's just not guaranteed.

I didn't have a lot of probs with ms but I went with brf out of 24 jars only two didnt germinate.

Thanks for all the replies, you guys. Here's some more info to my setup-

Its Rye grain, soaked overnight in the fridge, drained, PC for 60 mins at 25psi, inoculated into a self healing injection port then placed into a cardboard box to be dark and right now they are up in my pantry as high as I can get for a few extra degrees warmer.

The reason I used so much liquid culture from the spore syringe is because i shook the syringe and looked inside and could hardly see any spores. I could see 1 or 2 clusters inside. I don't know if it contained 1 whole spore print or only a scraping from one. So I wanted more spores in each jar. I think there werent much in there. (I realize the spores are microscopic, but I was looking for small dark clusters)

So right now I will have to be patient. This may be my last try for a while as I cant spend more money on spores right now :frown:

You should definitely not do this. Remember spores are sold for microscopy use only. Anything to do with cultivation should never ever be brought up with vendors. IF you plan on contacting your vendor be careful with word choice, and don't flame them. These things happen, and most vendors are very apt to helping their customers complaints.
