Tampanesis on Agar

Hi,

As part of an experiment, I have some P. Tampanesis spores on antibiotic agar, as I had previous problems with bacterial infiltration.

The recipe for the agar is the potato-based one with a pinch of fish antibiotic:

This strain come from Florida and where I live it is very dry (and not humid like Florida).
I was concerned that the agar might dry out.
It has also been 15-17 Degrees Celsius, which is not ideal temperature, as they require at least 20-25 to establish mycelium.

As the weather warms up, over the next month it should get to 20 degrees, but can anyone see any problems with the agar drying out if I leave them for a few more weeks, or whether I could add water to it if this is a problem?  At present I can see no activity and the spores have been there for about a week.  Here are two photos:





Are there any other suggestions or tips anyone could make?  Thanks

Those sure are some sedate looking spores you have there. Could be the problem is the cool temp. Maybe if you could get the heat up to 27-28 it might goose the process some. Anyway, that is what I would do, but I am not sure if the TC's would agree. In fact, I think they would probably say something like "keep it at 23-24".

Thanks for the feedback.  One thing I have noted with agar is its propensity to dry out as time progresses.  Can you see any issues in applying some PC cooked water to the agar plate over the flow hood?  Assuming there is no action after 2-3 weeks and that the spores do not drop dead?  Cheers

Thicker agar tends to dry out a lot slower. I had the same issue a while back

Maybe you're cover just isn't seal it well enough keep humid air from flowing out.

Might wanna try some no-pours in PP5's.  Haven't any issues with it drying out, though it's usually like 50 RH where I live.

I spawned some popcorn casings and had double-overlay cause I didn't put enough hydrogen peroxide in my automated aquarium mister.  I only got one mushroom so I cut off the head part where the seeds fall from and put it in a jar of LC and sprayed it all over a tin of PF cakes I made with gravel, cardboard, and bisquick in my microwave.  I think it will be good cause B+ is so potent.
Triggered yet?
