Alternatative tek for culture storage

Not sure if someone has posted similar before, but this is how I've simplified my culture storage for my gourmet ad medicinals. I use the standard culture tube method but instead of using agar I use grain instead. The grain I use is hard red winter wheat. Here are some pics so you can get an idea.



The pros for using this method is what made me switch. firstly when using the standard agar method you need to cut or scoop (whatever way you do it) a piece of the culture from the tube and transfer to start a new plate for expansion. with this method I just open the culture tube, remove the polyfill wad, and remove a single grain to transfer to a new agar plate for expansion onto grain. I like this method because it allows a set uniform size of culture to be transferred to the agar dish and basically contains hundreds of separate grains which basically translate to plates.
Hope this helps simplify the process for someone or allows a creative view that inspires others to experiment. Any questions are more then welcomed.
BTW I start the cultures from standard agar wedges from either a stored tube or dish.

i do a variation of that with little woodchips. I think with woodlovers they stay vital on it longer than on grain (mostly because it takes longer to digest and is less nutritious)

Could probably shove a piece of stick in there with the grains and maybe get the best of both worlds. (easy to extract grain and long lasting wood)

Well you all have a point. These tubes have been inoculated from traditional agar + popsicle stick culture tubes which I have as back up for proven long term storage. I haven't used these for longer then 2 years so I cannot vouch for it any longer. But I have used an agar petri for more then 5 years without any popsicle stick or any piece of wood. I think as long as you refrigerate and don't let your medium dry out, you'll be fine. Also there is half a popsicle stick in these grain tubes. The key is to transfer them into storage right as soon as they fully colonize. that way in cold storage they slow way down and have a long long time to continue metabolizing the grains. Also, hard red winter wheat are hard little grains - they take a while to eat them up fully.

There's a difference between a culture staying alive or staying vigorous and young.
And i would recommend not first letting it fully colonise before putting in the fridge, with slants it's also the practice to just let the wedge you have put in recover and then refridgerate, so it still has space to run in the cold.

That is exactly what I do. when I say fully colonized I am talking talking about the pre stage - when the first wispy growth is visible throughout the grain. Slants and agar are put into cold storage at about 80% for me. I leave a small rim around agar dishes. As per GGMM. I have never had a problem with any culture not being vigorous after that time. At least not anything worth complaining about. The true key is to store cultures that are young i.e. early division. The oyster you see in this thread will show you what I mean by "came back to life".

However, I must state the importance of starting from spores and getting a new culture started every couple of years. Also, let me make it clear that as time passes in storage cultures very slowly start to loose vigour - that is a fact. With proper storage you can expect good results over a few years time.
