Vacuum-sealing your PDY
    #2211322 - 12/31/03 09:12 AM (13 years, 11 months ago)

Sorry if this is the wrong forum, but noone really uses septum bottles....

Shit.... Last night I was trying to propogate this culture syringe (Shiitake -- my last one, damnit) and I was using these new septum-bottles I have and I didn't realize I was going to vacuum-seal them if I didn't let them cool before sealing them. Well, when I inoced them the plunger pulled down by itself, and figured from Boyle's Law there should be like ~26 cc's of air left in the jar, down from 400. I tested a control jar with a 30cc syringe and emptied ~ 30cc X 13 = ~390cc's of air missing (this isn't exact.) This is like almost all the air in the jars.

Now, I didn't want to suck in unsterile air, so I left them like that in a rather cold room. This shouldn't be a problem for one day, right? I mean it doesn't have a brain so it can't suffocate, right? I guess if they last in the culture syringes....

I hope they're going to be o.k. because that was my last syringe of shiitake and because of the vacuum it sucked all of it up so it's all in the PDY now. I figure I can put them in a bubble and spray paracetic acid, seal and let it break down and open them in the bubble, but not until tonight....

I guess from now on I'll let them cool in the PC.

Thanks!
