To add onto this, im working with a Spore Print, so im going to have to briefly open the caps of the jars to put the spores in.
Anyone have any information or experience regarding that?

It's gonna end bad. Go to agar first.

It can be done, however, you have to make sure the spore print is not contaminated. It's hard to tell in a lc.

Agar first!!!
Especially if this is a wild print you are working with. If so, it is almost guaranteed to be contaminated.

Print to LC is a bad way to go, way too much risk. Get clean germination of the spores on agar first, then transfer a wedge of clean mycelium to the LC.

What if the print was made from cultivated fruits in a sterile environment?

It wasn't a sterile environment.

How did you have a jar fall over in the PC?
Plates: http://www.ebay.com/itm/Sterile-Polystyrene-Petri-Dish-Plate-100x15-mm-500-cs-/111668743267?pt=LH_DefaultDomain_0&hash=item19fff9cc63
Agar Agar: http://out-grow.com/culture-media-isolation-tools-c-10/agar-agar-powder-p-114.html
If you have grain water from some soaked grains, that's all you need in terms of ingredients. If you don't have any grain water, then you should also grab some light malt extract. http://out-grow.com/culture-media-isolation-tools-c-10/8-ounces-of-light-malt-extract-p-79.html
You need a SAB\flowhood, and empty liquor bottle\suitable agar vessel, a scalpel\xacto knife, a torch for flame sterilization and then some cling wrap or parafilm to wrap the plates in, then you're set.

The word "sterile" does nothing but cause arguments, you could sterilize grains at 60psi for a year and someone would question the sterility. Some would argue that a sterile environment is an impossibility so the discussion ends there.
You can grow and have mushrooms drop spores in unusually clean ways though. Transfer supposedly clean growth to supposedly sterile subs in small jars and let them fruit invitro. When the shroom matures tip the jar on its side and let spores drop, now inject sterile water and draw out spores. So the jar is never opened once. This liquid may mix with substrate and so have nutrients which mix with the spores. There may be methods to overcome this but if you are not a vendor it might not bother you.
I suspect some of the vendors are using techniques unknown to the general public. Some people talk as though they are all pretty much identical.

hiperhiper said: so what's the right procedure with grains? I've checked the teks and nobody said anything about this. I didn't know I was not supposed to use syringe on grains. What is cleaning up syringe on agar? what does it mean?
