Deep agar dishes

Ok so im using 1/8th or 1/4 pint jars for my agar dishes just cause i was at the store and well it was right their so i tried it out.. First jars i let fruit so i could take a pin an the pins i transfered from. The first agar jars are very healthy. now since the top is colonized im wanting to drop wedges in wbs jars an make more transfers to keep going. My problem is that my agar is about 1/4-1/2 in. Thick
Being that i only need the mycelium can i cut it an possibly scrape what agar is stuck to the bottom of said cut piece

yea u can.  be careful though.  maybe use a different tool/spoon to kinda scoop it off the top.
if its a 1/4" i'd just do standard.  u can do standard drop with 1/2" too, just might take a few more days leap off wise

Sweet. At least theirs hope hahaha. I tried letting the first colonize all the way but the agar eventually dried out by the time the mycelium was penetrating to the bottom.

It's not supposed to penetrate to the bottom. We use agar as a 2D surface so we can spot contams.

Ohhhhhh well fuck me that makes total since lmao :blush: i was sittin here thinking well i wonder if i should wait longer to let it penetrate more but thats a no. Thanks 36fucking5.. Also one of your post about hidden contams saved my ass some work recently. Its nice that others have ran into unfortunate problems and found a solution. Thanks again lol

One of the nice things about doing glass petris or Pasty plates is that you can just mix agar in open air, fill the containers in the comfort of not an SAB, and then cook it. I think that makes it easier to not overfill - the fact that you can work unencumbered by sterile procedures.

I also don't have problems dropping thick wedges- Pasty plates tend to have thick edges because of the shape of the bottom of the containers. The myc will colonize the edge as it makes its way to the grain.

I'd drop a thick wedge before I'd scrape the myc, personally. Less steps, less vectors :thumbup:

I use the 1/2 pint Kerr's all the time. They work great, never any condensation issues, easy to see through and really easy to prep. The edges are much more thick than the center but it hasn't been a problem for me(yet). If you're not doing a ton, I think they're the way to go.

Yea i love how i can simply mix an jack with water content an such before it gets sterilized. I got overwhelmed reading everything about pouring hot agar an such. I dont see why more people dont use this method.. If my dumbass can get it to work then anyone can haha

It's because pre-poured plates are more likely to have condensation, and the containers you need (glass petris don't actually work for no-pours, anyone who says otherwise hasn't tried it) are terrible for trying to see through.

Pouring petris isn't any harder than doing G2Gs and you can be more consistent.
