Update at bottom on myc/bac??? fight.
    #2207897 - 12/29/03 04:21 PM (13 years, 3 months ago)

A friend of a friend recently innoculated 3 creeper quart jars and 3 mazatapec jars (millett based). She said jars are about 80-85% colonized. This friend is pretty sure that all of the jars are at least in the beginning stages of catching bacillus (sour odor detected she said) though he/she says that there are no visible brown spots characteristic to this contam. She/he wanted to know if it is possible for the mycelium to fight it?

MAIA, the friend also believes that bacillus is not airborne (she/he is using 2 HEPA filters anyway and has done several successful experiments, never been contamed before). So it seems unlikely this is an airborne contam.

My friend's friend does not want to ever have to have spores shipped again, so what should I tell her would be the best way (if there are any ways) to try and save some of the mycelium to make more spawn? They are in quarts now, she said she thought about cutting healthy looking pieces off and soaking in peroxide and doing a G2G in 1/2 pint on millett. Worth a try?

Thanks for the reply.

you can just make a spore print, or you can cut away a chunk of that mycelium and put it in DI water in the dark, in a sealed container in the refrigerator for an indefinate amount of time. I would reccomend some 20:1 water to H202 or 100:1 water to bleach. If you want to start another spawn run, cut away a chunk of mycelium and spawn something new but its not really worth it to try to spawn something with a ratio of more than 5:1 spawn to substrate.

she could always do some agar work with the healthy mycellium and then use that to innoculate down the road.

I have had a lot of experience with Bacillus and have never once seen the mycelium win.

> MAIA, the friend also believes that bacillus is not airborne

Bacillus forms endospores when it dries out.  These can become airborne and spread the contamination.  As soon as an endospore rehydrates the bacillus will start to multiply.

> So it seems unlikely this is an airborne contam.

What it seems and what it is can often be different.   (Anthrax is a good example of an airborne bacillus used as a weapon.)

Bacillus is also mobile.  The stuff will move around no its own from one location to another.  This is a double hit... if it is dry, you get airborne contamination... if it is wet, you get the stuff sliming around.

Thanks for the input everyone. My friend decided to do agar work (seems successful at this point so she says) with the jar which seemed the worst. She/he also separated 2 jars that seemed to not be in good health from the 2 that he thought might survive. My friend checked on them a couple days ago and to her suprise, the jars seemed to colonize to about 95%! No more bad smell either ... ?

I told her I would ask about this and see if anyone had any idea what may have happened. He/she also reported to me that the jars in the incubator reached 100%.

Thanks!

Does this mean 3% H2O2 Solution (like lysol) should be dilluted with
water 20:1 ? Or is should it be 100% H2O2 dilluted with water 20:1 ?

I assume you mean the 3% , but before i am doing something wrong, i thought better ask first

Sorry , but lysol is not h2o2 . Go with bleach , 1 part for 100 of water . Maybe a solution of diluted lysol could work , but hippie3 has great success with bleach !

Oh,  I'm not sure its work with bacillus , so ... :bong:

It's 1 portion of 3% h2o2 solution to 10 portions of water (1:10). 1:20 is a weaker solution but should have a purpose depending on the situation, anyway 1:10 is still safe.
