syringe sterilization

Hey everyone, i just have a quick question. i'm having problems with my pf jars lately, they get bacillus every time now. I've been reading some teks and noticed that I've been flame sterilizing the needle, but then i wipe it with alcohol. I'm wondering how this will affect my jars. Would it have anything to do with them all getting wet spots and going green? Any input is appreciated, thanks guys.

they ALL got contaminated? That sucks, especially since the error could have occured at any stage, maybe you didn't sterilize, maybe you didn't presoak. Alcohol and flame is usually enough, you MIGHT have gotten a bunk syringe, the only way to know if you've got clean spores is to leave a jar sterilized but NOT inject it. If the rest go green and that one stays clean I'd say its the syringe, if they all go contam again its your sterilization technique.

ipa and flame is enough, maybe the syringe is contaminated though, like the spore solution itself not the needle, also how's the water content in the jars? maybe you have to much water in them

they all go bad. never happened before. i've made my own syringes. presoaked 12 hours at least. i've done everything i can to be sterile. the water was boiled, used lysol before doing anything, even pc'd the jars for 1.5 hours. the only thing i'm not sure about is the spores. they're only a couple of months old, and I've kept them in a non drafty, cool area (no moisture) I can't think of anything else. I see mycelium growing, but the green quickly takes over. Would bad spores cause wet/green spots? also, i use less than 1cc of solution per jar.

how many batches have failed in this way? Have you had any successful jars w/ this syringe, have all the bad ones been w. this syringe?

About 4 or 5 batches turned out this way. all different syringes.

well maybe contams are entering the jar AFTER innoc? How are they sealed and incubated? By the way I REALLY like making prints in a still air glovebox! Load the syringes with steril water, clean yer materials, and away you go! with no contams! I like to boil distilled water and suck it into a syringe. The only thing I use to sterilize the needle with is alcohol and lysol. dk

Just a thought , perhaps it would not hurt to a inject a karo jar along with your bfr-s that would tell you if your print is contaminated as long as u have cleaned the syringe via the boiling water method . Just a thought ?

pf for simple minds. verm on top, yes, ziplock, yup, fridge? no. but i can give it a try. incubated in double rubbermaid with fishtank heater.i do boil my water, then leave it overnite to cool, then make my syringe (shotglass method), and leave sit for at least 12 hrs b4 usage. the karo, i think i could try that, though i've never had a bad experience with everything i've used. thanks everyone.. i hope i can get this shit cleared up with your help. ?buonas noches!

"i do boil my water, then leave it overnite to cool, then make my syringe " that may be your problem,try boiling your water for 10 minutes them turn the heat off and fill your syringes as soon as it stops boiling (a couple of seconds) then leave the syringes to cool. leaving the water over night to cool in the pot gives contams plenty of chance to get in.

yeh thats the way I do mine syringes too. Again try investing a glove box I made mine for 8 bucks in under 20min.
I say clean out that incubator like CRAZY. Lysol the fuck out of it, and start with new shit.
HEY OOKLA! what is gentamycin, and what does it do for yer syringes?

Gentamycin is an antibacterial that can withstand PCing, it wont help much with the green but it will prevent the bacillus. Peace - @cro

no I don't have a tek. I just found a box and some gloves I liked, cut holes with a hot knife and taped it all together.
for some teks try the FAQ's I havent looked in there for a gb tek, but you might find one....
use the search feature for "positive pressure glovebox" or "still air glove box" that should give you some ideas.
