11 days passed - grain jar

Hey guys, This is my second time after my first attempt that failed. I Have 3 quarter jars with Rye Berries. I've inoculated on Sunday July 26th. I waited 4 days and two of them had good growth, I shaked. then 2 days later I had more grow and I shaked again. I could only shake the 3rd jar today (after 11 days). After you shake some of the mycelium dissapear I've noticed.



2 questions:
1-How long do I still need to wait ? I still see that 2 jars are not 100% colonized.
2-Am I seeing a minor contamination on this one (top)?

Top 2 look contaminated. Bottom one is probably bacteria.

How did you inoculate the jars, MS or an agar wedge? They do look bacterial, because the mycelium doesn't seem to have a strong hold on the grains.

I've used a spore syringe from a vendor that I bought (the same one that I had a problem before).
Is it possible that the syringe content is infected?
I really don't know what I've done wrong, this time I purchased those jars from the internet which were already sterilized / PC , they have self healing spots and I injected inside a SAB box which I cleaned with lysol (it's all plastic and it's sealed, I also cleaned the needle before injecting. The needle was being stored in my fridge.

Syringe to grains is a failure waiting to happen. More than likely the syringe is dirty and needs to be cleaned up on agar, And I wouldn't trust pre sterilized anything

so what's the right procedure with grains? I've checked the teks and nobody said anything about this. I didn't know I was not supposed to use syringe on grains. What is cleaning up syringe on agar? what does it mean?

An MS syringe is not sterile, so contams are almost always present. That's why it's recommended to drop them on agar first to see them, and only transfer healthy grow to your grain jars.

That's the next problem. Do not trust online vendors that sell grain jars. I made this mistake my first time getting started and the grains were very dry when they arrived at my doorstep. I injected my syringe in an SAB, but there was no growth after two weeks.
It is 10X better to make the jars from scratch, properly sterilized in your own PC, than to rely on someone else to do it for you, not to mention paying a large sum of money for something you could make for a fraction of the price.

ok but what if the agar gets contaminated?
how do I really start clean from the first step?

Shake after the jar looks ~30% colonized, then shake again when the jar gets to 100% and you're ready to spawn (obviously lol).
