Jar lids polyfill problem
    #22040963 - 08/04/15 07:27 AM (8 years, 5 months ago)

Hey guys I'm going back in after a small break ,I did 20 rye berry jars about 2 weeks ago and nothing is happening . I am worried that the polyfill is the cause . I think I'm using a different one , I made it as tight as possible like usual but this time after the pressure cook the polyfill looks cooked kinda it's all stiff almost cracky.

My jars and grains are perfect nothing burnt ,there was still water in the pc after the 2 cook. Moisture content is perfect the aluminium did his job

Not sure what is going on  could I pc some new lids and do the transfer in a sab?would it be worth it?

What did you use to inoculate?

Two weeks seems suitable for MS germination
Two weeks does not seem suitable for LCs,g2g, agar, ect

It may not be a polyfill problem

Its a MS inoculation but still I find it pretty weird that there is nothing after 2 week all my prior grow was starting after 3-7days now it's been 12 days and absolutely nothing. I heard that after 2 weeks you should throw it if there is nothing happening

The only thing that seems different is my polyfill

I dont use polyfill for this reason. The only contaminants I ever got were from jars with poly as a breather.

i think even if ur poly was mangled by the PC heat, u'd still at least see some germination, but probably would stall cause no/little GE.

maybe shake it and see if u see any growth hiding in the middle.

how big is the hole u stuffed the poly thru?

All my past attempt (around 100 jars) were successful with poly fill  :ooo:

The holes are 1/4 stuffed really good .

If you say I should see growth anyway maybe I'll just wait I guess .maybe it's my syringes too, they are like 6 months old stored in a box in my basement

yea, poly works fine, but like u said, some brands are weird
i think some people use larger holes for poly (frank for example), cause its easier to stuff them.

I'm only guessing on germination, because in the past, i have used LC's with no GE, inoc with spores, and it worked, so i would guess same thing with grains.
also, spore syringes can sometimes spontaneously germinate, and there is no GE in that.  so my first guess would be its the spores

I think its easier to stuff smaller holes with poly, at least when you want it so damn tight.

but yeah I'm with leaf you'd still see germination even with 0 GE, if there's nothing there to breathe yuo dont need ge anyway right.
