Good idea to leave jars in glovebox to avoid contams?

I knocked up a few jars of liquid culture in a large glovebox.  I took a good sized tub, put it on the side, covered the opening with saran wrap with a flap to work out of, which will fold closed.  I sprayed the interior with disinfectant, let it sit for awhile, then got to work.  I left all the jars in the box while I was working and I thought, why not just close the flap and put the box on a shelf.  Seems like it would be a lot safer in that pre-cleaned environment than in the cupboard.

Any thoughts on whether this is a good tactic or not?

I had the exact thought, until I realised that I need my SAB/glovebox for more work, and that my jars have filters on them for a reason :smile:

Yes, you could and I agree, theory sounds great... the problem is you just lost your glovebox. It just became an incubator haha. If it's your only project than go ahead but if the jars are securely closed and relatively disinfected before they're reopened then there's no reason to leave them in there.

Thanks.  I'm not going to need the glove box again until the LC is ready anyway so its not a problem losing its use for awhile.

I've had bad luck with contamination, 40% loss ratio my last two tries (I'm new at this, today was my third attempt).

Does contamination usually occur at inoculation or before, or is it something that happens during colonization?  I know it can be both, just wondering which occurs most often.

Well as long as you properly sterilized the spawn or cakes then it's obviously more likely to come post-inoculation.

Any time it's opened it can get contaminated. So if it's properly closed with a good filter it theoretically shouldn't contam just sitting there for a while. Bacteria and mold spores usually get in during transfers either during inoculation/g2g/spawning/etc.

And I need to say, 40% isn't bad at all. I don't know about you but I'm not a professional grower so on a small scale (no $500 equipment) i can afford to lose my regular 20% without worry.

Used GT spores in half filled pint jars.  Half are polyfill filters half are not filtered, just sealed with silicon.
