When storing these plates it's fine to just have them sitting up right?
Or flipped over?

Unless your plates are pooling with water I see no reason to store plates upside down :shrug:

Hey pasty, I have 4 old colonized grain jars that have a few small area of bacteria. I cased them with coir/verm and stuck them in a mini mono. I never messed around with invitro, I just didn't want to spawn them to bulk and waste my time. Anything special I need to do with them?

I bottom water once the pinset is in. Otherwise mist if they look dry. Thats all there really is too it.

I am also giving this a whirl, except using wide mouth half pint jars and these liquid culture lids

Kinda just thought of the lid thing last second because I had extra, I'll report on how it works.

I used 2g LME to 2 g agar with 100 mL distilled water.


I also have some end of the boil cycle water frozen, was wondering what you thought about ratios to agar, or if you have any experience with this?

Just use the grain water as is, I usually don't bother to dilute when using it for agar.

im bottom watering from now on. with the summer heat misting has been causing some issues on dead aborts in tubs for me lately. ive recently been pulling back the liner, then running the sink in it and letting the substrate float for an hour. dump off put back into fruiting.

need to make sure to save my next soak for the liquid cultures and the plates now.

man i have way too many pasty plates now and not enough jars to fill. ill have to crack open the bags i think. or do some a2g from hell to use up some of these plates. I feel like im  back in the stone age doing A2G. its like slow motion or something.:tongue:

I have tried bottom watering bulk but it always gets water locked.  To far for it to travel and too much pressure from the water already in the bulk. I find its less an issue with higher spawn ratios tho.

You will end up with a buttload of soak water once you start freezing it. I even label the type of grain it came from.

Do a tiger drop into a master and shake the hell outta it. The master will colonize very fast with that much agar and all the culture rubbing on the grains.

yeah i need to do that again. maybe 10 of them. Oh I got AA plus, Peu, pe, ape coming to a threatre near you. also burma clone, syzygy clone 1 and 2, pf classic clone, tc clone. the aa plus multispore syringe is clean. it went on a plate a few days ago and germiated already.


but then i have to g2g after that. i like doing 50 jars in one sitting with some LI or slurry. its like pouring people a beer only maybe a little easier. i get more for my money with the liquids rather than g2ging the quart to 10 quarts to get 6 day colonization. plus i can do it all in one sitting so it makes the clean work super easy and efficient. only so much time working 60 hours and doing the hobby for fun.

When transferring the spores over to the agar, 1) can I use a syringe a put a tiny droplet in the center of the agar? Or 2) use a sterile swab to transfer spores from the spore prints. Will both of those work?

Living in Europe I'm having trouble finding small and cheap enough plastic jars for this propose. IKEA is a possibility, so is ordering a Glad mini round pack from the UK or US, so is using petris of 100% polystyrene which is by far the cheapest.

Yes both those methods will work.  Might as well just let the drop fall on the center of the plate, so it has some moisture to germinate on.
