Hi there,
Can anyone explain me this? With the same grain, sterilized 3 hours. I transfered Agaricus blazei to two Jars and Agaricus Hortensis also to two Jars. Agaricus blazei mycelium populated all the grain on jar sucessfuly no contams, but Agaricus bisporus, always contaminates, i can se everytime three or four grains starting with early trichoderma. I dont understand? Why didnt blazei contaminated with the same grain? I find extremly hard to grain transfer agaricus bisporus hortensins always getting trichoderma ... Any idea? tips? Sugestions?
Thank you all

Maybe the bisporus culture is contaminated?

But i saw the contaminated grains, a few of them, they were not even colonized and still not touched by the mycelium and they were already starting to get green?

still sounds like your culture is contaminated even if you can't see it. or maybe you didn't clean the jar or whatever as much.

Some grains itself started to color green, so I suspect the sterilization procedure was not enough, I used a pressure cooker for 3 hours, but nevertheless I used BIG 5 liter jars filled with grain with 1/3 space free. I suppose 3 hours is not enough for this 5 liter jar spawn? Advice please?

no, trichoderma spores die around 140F so bacteria are usually the only things that survive the autoclave cycle.

Why use a 5 Liter jar? Seems a bit silly to me
I would think the inside of the grain jar is not being sterilized due to the insulation caused by the other grains. Try a 1L jar if you have one and do not inoculate, wait a week and see if trich grows. If it grows nothing then it is either your culture or the large size of the jars.
Post in Gourmet/Med next time, you get more responses and we try to keep support-type questions out of advanced.

ToadStool, NICE great idea, i always thought that trich only appear if mycelium is there, new bie thought of course, this way i can get this right and see what is the root cause of it, and i dont need to waste mycelium. Yes it could be also due to the insulation.
Thank you very much for insights, im going to do this tests.

Blank jars cost a little bit of supplies but they allow you to test your sterility pretty well. Im growing in a new house in the bathroom with a new PC that only goes to 12psi so i have done a few blanks to check my work. i got the idea from the vets here
